摘 要:目的:研究活血降糖饮对长期高脂饲料喂养大鼠糖、脂代谢的影响及其分子机制。方法:采用长期(28周)高脂喂养建立胰岛素抵抗、高脂血症大鼠模型,将模型大鼠按随机数字表法分为模型组、中药组(活血降糖饮)、贝特组(非诺贝特、阳性对照),另设正常对照组。药物干预治疗后行口服葡萄糖耐量试验(OGTT)及胰岛素释放试验(IRT)并测定各组大鼠血脂及相关蛋白表达、肝组织病理学检查及肝脏甘油三酯(TG)含量。结果:与模型组比较,中药组大鼠OGTT、IRT明显改善,血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、游离脂肪酸(NEFA)明显降低;而高密度脂蛋白胆固醇(HDL-C)、肝组织胰高糖素样肽-1受体(GLP-1R)蛋白、肝组织、脂肪组织葡萄糖转运体-4(GLUT-4)蛋白表达较模型组明显升高,肝脏脂肪沉积减少,TG含量明显下降,脂肪组织GLP-1R蛋白表达与模型组比较差异无统计学意义。结论:活血降糖饮可有效治疗大鼠高脂血症及胰岛素抵抗,其作用机制可能与减少肝脏脂肪沉积,增加肝组织GLP-1R、GLUT-4及脂肪组织GLUT-4蛋白表达有关。Objective: To study the molecular mechanism and effect of Huoxue-Jiangtang prescription on regulating glucose and lipid metabolism of hyperlipidemla and insulin resistance rats. Methods: The rat model of hyperlipidemia and insulin resistance was established by feeding a high-fat-diet for 28 weeks. The model rats were randomly divided into model group, medicine herb group (Huoxue-Jiangtang prescription) and fenofibrate group. Corresponding therapy were given through gastric tube for rats in different treatment groups. Meanwhile a control group was set up in normal animals. Oral glucose tolerance test (OGTT), insulin releasing test (IRT), serum lipid level, and lipid metabolism related proteins were detected. Liver pathology was determined and liver triglyceride (TG) content was examined. Results: Compared with model group the OGTT, IRT, serum concentrations of total Triglyceride (TG) , cholesterol (TC) , Low-density lipoprotein- cholesterol (LDL-C) , non-esterified fatty acid (NEFA) in medicine herb group were decreased. At the same time, serum levels of high density lipoprotein-cholesterol ( HDL-C), glucose transporter-4 (GLUT-4) protein in liver and adipose tissue in medicine herb group were increased as well as the expression of glucagons like peptide-1 receptor (GLP-1R) protein in liver. Meanwhile, liver lipid accumulation and TG content were decreased in medicine herb group. Conclusion: It suggested that Huoxue-Jiangtang prescription is effective on treating hyperlipidemia and insulin resistance in rats. The mechanism may be related to inducing the expression of GLUT-4 protein in liver and adipose tissue as well as increasing the expression of GLP-1R protein in liver.
