摘  要:合成了1种新型具有良好生物活性的4,5-二甲基-3-腈基-2-呋喃胺对苯二甲醛Schiff碱(FTS).并通过红外和氢核磁共振谱对FTS的结构进行表征.应用荧光光谱法、紫外光谱法研究了该化合物与牛血清白蛋白(BSA)之间在不同温度下的相互作用.实验表明,FTS能强烈猝灭BSA的内源荧光,猝灭机理为静态猝灭.在此基础上计算了二者相互作用的结合常数、结合位点数及热力学参数等,结果表明,FTS与BSA分子以摩尔比1∶1结合,其结合反应主要是熵驱动,主要作用力是疏水力.同时,应用同步荧光考察了FTS对BSA构象的影响.A new compound with good biological activity 4, 5-dimethyl 3-cyano 2-Furan amine terephthalaldehyde Schiff base (FTS) has been synthesized. The structure of the FTS has been characterized by infrared and hydrogen nuclear magnetic resonance spectroscopy. The interaction between FTS and bovine serum albumin (BSA) was investigated by Fluorescence spectrometry and ultraviolet spectroscopy at different temperatures. The experimental results showed that the FTS can strongly quench intrinsic fluorescence of BSA and quenching mechanism is static quenching. On the basis , the binding constan and the number of binding sites and thermodynamic parameters of the interaction between the two have been calculated. The research results indicated that FTS bound BSA with molar ratio of 1 : 1 and the combined process is mainly driven by entropy, interaction is mainly hydrophobic interaction. At the same time, the effect of FTS on the conformation of BSA was analyzed using synchronous fluorescence spectroscopy.
