摘  要:为探讨乙二醛(GO)和甲基乙二醛(MGO)对上皮细胞的细胞毒性及其机制,分别以MTT法检测两种活性羰基化合物(RCS)对ECV304细胞活力的影响,评价其细胞毒性;DCFH-DA探针法检测氧化应激;用罗丹明123检测线粒体膜电位及线粒体形态变化。结果表明,GO和MGO对ECV304细胞的IC50分别为3.6mmol/L±0.1mmol/L和1.3mmol/L±0.1mmol/L;氧化应激水平小幅度增高,线粒体膜电位随发生改变;线粒体形态发生改变。GO和MGO的细胞毒性与氧化应激水平增加相关性较低而与线粒体损伤相关。To explore the cytotoxicity and its mechanism of GO and MGO on epithelial cells,the cytotoxici-ty of two reactive carbonyl species were measured by MTT reduction assay.Oxidative stress was detected by a non-specific ROS probe (DCFH-DA),and the MMP and mitochondrial morphological changes were detected by using rhodamine 123.The IC50 of GO and MGO on ECV304 cells respectively are 3.6 mmol/L ±0.1 mmol/L and 1.3 mmol/L±0.1 mmol/L.The level of oxidative stress had a slight increase while the mitochondrial membrane potential was alterative,and mitochondrial swelling occurred.In conclusion,the cytotoxicities of GO and MGO were mainly due to the mitochondrial dysfunction,and showed weak corre-lation with oxidative stress.
