摘  要:采用石油醚-硫酸溶液两相溶剂水解,UPLC-PDA法测定白花延龄草根茎和果实甲醇提取物中薯蓣皂苷元含量。色谱柱Waters BEH C18(2.1mm×100mm,1.7μm);检测器PDA。流动相乙腈(A)-水(B),线性梯度洗脱:0min^8.0min,50%~90%A;8.0min^8.1min,90%~100%A;8.1min^11.0min,100%A;11.1min,100%~50%A。流速0.4m L/min;柱温50℃;检测波长192nm;进样量5μL。结果表明,白花延龄草根茎甲醇提取物中薯蓣皂苷元含量为0.03%,果实甲醇提取物中薯蓣皂苷元含量为0.16%。With petroleum ether and H2 SO4 two-phase solvent hydrolysis ,Determination of diosgenin in the rhizome and fruit methanol extract of T .kamtschaticum Pall .using UPLC .chrom column:Waters BEH C18 (2.1mm × 100mm ,1.7μm );Detector:PDA .The mobile phase with a linear gradient elution of acetonitrile (A)-water(B):0min-8.0min ,50% -90% A;8.0min-8.1min ,90% -100% A;8.1min-11.0min , 100% A;11.1min ,100% -50% A .Velocity :0.4mL/min;The column temperature :50℃ ;the detection wavelength:192nm;sample size:5μL . The results showed that ,the content of diosgenin rhizome methanol extracts was 0.03% ,and the fruit methanol extracts was 0.16% .
