摘 要:目的:观察蝉蜕、僵蚕对系膜增生性肾炎模型大鼠的治疗作用及对肾组织Toll样受体4表达的影响。方法:选用清洁级Wistar大鼠60只,随机分为正常对照组、模型组、蝉蜕高剂量组、蝉蜕低剂量组、僵蚕高剂量组、僵蚕低剂量组,采用改良慢性血清病法复制Ms PGN模型,各组大鼠分别给予蝉蜕、僵蚕不同剂量颗粒冲剂灌胃,正常对照组和模型组给予等量自来水灌胃。检测各组大鼠24 h尿蛋白定量,血清ALB、BUN、Cr、TC。HE染色观察各组肾组织的病理变化;免疫组化法观察各组大鼠肾脏组织中Toll样受体4的表达情况。结果:蝉蜕、僵蚕各剂量组大鼠尿蛋白明显减少,血清白蛋白不同程度升高,肌酐、尿素氮降低。肾脏病理改善,肾脏组织中Toll样受体4的表达减少。结论:蝉蜕、僵蚕能改善脂质代谢,减少蛋白尿,抑制肾小球系膜细胞的增殖,减轻系膜基质积聚,其作用机制可能与抑制肾脏组织中TLR4过度表达有关。Objective: To observe Periostracum Cicadae and Bombyx Batryticatus in treating mesangial proliferativeglomerulonephritis rats and influence of kidney Toll - like receptor 4 expression. Methods : 60 Wistar rats of clean gradewere randomly divided into normal control group, model group, high dose group of Periostracum Cicadae, low dose group ofPeriostracum Cicadae, high dose group of Bombyx Batryticatus,low dose group of Bombyx Batryticatus. The MsPGN modelwas copied by the method of chronic serum disease improvement and the rats were given the Periostracum Cicadae, Bom-byx Batryticatus of different doses of decoction orally. The normal group and model group were given equal volume of water. We detected the rats ' 24 hours urine protein, serum albumin, blood urea nitrogen, creatinine and total cholesterol.Pathological changes of renal tissue were observed in each group by HE staining. Toll - like receptor 4 in renal tissue wasobserved by immunohistochemistry. Result: Urine protein of rats in each treatment group decreased significantly, serumalbumin increased at different degree, creatinine, urea nitrogen decreased. Renal pathology improved, the expression ofToll like receptor4 in renal tissues reduced. Conclusion: Periostracum Cicadae and Bombyx Batryticatus can reduce pro-teinuria and inhibit glomerular mesangial cells proliferation and mesangial matrix accumulation. Its mechanism may be re-lated to the inhibition of TLR4 excessive expression in kidney tissue.
