摘 要:目的:建立黑面神茎中总黄酮的含量测定方法,优化其水提工艺参数。方法:采用紫外分光光度法,以芦丁为对照品,Al(NO3)3-Na NO2-Na OH系统显色法,测定波长511 nm。以总黄酮含量为指标,用正交试验考察提取时间、固液比及提取次数对总黄酮含量的影响,优化水提工艺条件。结果:芦丁在9.0-52.0μg/m L浓度范围内与吸光度呈良好的线性关系,r=0.99935(n=7),回收率为99.1%,RSD为0.83%(n=6)。各因素对总黄酮含量的影响程度为:提取次数﹥固液比﹥提取时间,最佳水平为A2B2C2。结论:此方法操作简单,结果稳定,重现性好。水提最佳工艺条件:药材加20倍量水加热提取2次,每次1 h,经优化后总黄酮含量达38.58 mg·g-1。Objective : To establish the determination method for total flavonoids in stems of Breynia fruticosa and to op-timize the water extraction process of total flavonoids in stems of Breynia fruticosa. Method : Rutin was used as referencesample by UV spectrophotometry and 5% NaNO2 - 10% A1 ( NO3 )3 - 10% NaOH as color - developing agents and themeasurement was conducted at 511 nm. With total flavonoids as index, the extraction parameters including extractiontime, ratio of solid to liquid and extraction times were confirmed by orthogonal design. Result : The absorbency and concen-tration of rutin had good linear in the range of 9.0 - 52.0 μg/mL, r = 0. 99935 ( n = 7). The recovery rate was 99.1%,RSD was 0.83% (n =6). The order of factors was extraction times 〉 ratio of solid to liquid 〉 extraction time,and thebest match was A2 B2 C2. Conclusion:This method operated conveniently with stable result and good repeatability. The op-timal water extraction technology was as follows:ratio of solid to liquid at 1:20 (w/v), extraction 2 times and 1 hour foreach time. The total flavonoids in Stems of Breynia fruticosa was 38.58 mg · g-1 .
