针灸预处理对大鼠心肌梗死心肌S100A1蛋白表达的影响

摘　　要：目的:探讨针灸预处理对大鼠心肌梗死后S100A1蛋白表达的影响。方法:建立假手术组(NS组)、大鼠心肌IM模型(IM组)、心肌IM+针灸预处理组(IM+P组)。利用TTC法检测大鼠心肌梗死面积、TUNEL法检测心肌细胞凋亡情况,ELISA法检测MMP-2、TIMP-2、VEGF、S100A1等指标,利用免疫组化和Western blot检测心肌S100A1蛋白的表达水平。结果:TTC染色提示IM+P组梗死面积较IM组减少,TUNEL结果显示,与NS组相比,IM+P组和IM组细胞凋亡明显升高(P<0.05),且IM+P组明显低于IM组(P<0.05)。较IM组相比,IM+P组大鼠MMP-2、S100A1明显下降,TIMP-2、VEGF表达升高。免疫组化和Western blot结果显示,与NS组相比,IM组和IM+P组S100A1的表达水平明显升高(P<0.05),且IM+P组明显低于IM组(P<0.05)。结论:针灸对心肌缺血再灌注损伤起到保护作用,其机制可能与通过抑制S100A1蛋白的表达有关。Objective ：To investigate the effects of acupuncture pretreatment on SIOOA1 protein levels in acute myocardialinfarction rats. Methods ： We established normal group, IM group and IM ＋ acupuncture pretreatment （ IR ＋ P） group. The myocardial infracted area was detected by TFC method. The myocardial apoptosis was detected by TUNEL. The plasma levels of MMP -2 ,TIMP -2 and S100A1 were analyzed by ELISA and the protein levels of S100Alwere analyzed by im- munohistochemistry and Western blot. Results ： Compared to IM group, the infracted area and the apoptotic rates of IM ＋ P groups were significantly decreased （ P 〈 0.05 ） and the plasma levels of MMP - 2 and S100A1 were decreased and the plasma levels of TIMP - 2 and VEGF were increased. Immunohistochemistry and Western blot assay showed that S100A1 protein levels in IM and IM ＋ P groups were higher than those of NS group （ P 〈 0.05 ） and IM ＋ P group＇ s was lower than that of IM group （ P 〈 0. 05 ）. Conclusion ： Acupuncture pretreatment protects against acute myocardial infarction of rats by regulating S100A1 expression.
