摘 要:目的:观察赶黄虫草胶囊对复合因素致大鼠肝纤维化的防治作用。方法:选取SD大鼠100只,除空白对照组10只外,其余大鼠采用每周2次皮下注射40%CCl4花生油溶液,同时喂予高脂低蛋白饲料,隔日以10%乙醇为饮水等复合因素刺激制备大鼠肝纤维化模型,连续8周。造模4周后将大鼠随机分为6组,分别为正常对照组、模型对照组、鳖甲软肝片阳性对照组(灌胃给予鳖甲软肝片800 mg·kg-1·d-1)、GHCCC高、中、低剂量组(灌胃给予GHCCC 800、400、200 mg·kg-1·d-1)。给药8周后,各组大鼠取血、取肝脏,酶联免疫法测定血清Ⅲ型前胶原肽(ⅢPC)、层黏连蛋白(LN)、透明质酸(hyaluronic acid,HA)含量;化学比色法测定肝组织羟脯氨酸(Hyp)的含量;肝组织病理切片行Masson染色,镜下进行肝细胞脂肪变性及肝纤维化分级。结果:GHCCC高、中和低剂量组的血清ⅢPC、HA、LN水平和肝组织纤维化面积显著低于模型组的相应值;高剂量组肝组织中羟脯氨酸水平显著低于模型组;肝脏病理损伤明显减轻,其中高、中剂量组肝脂肪变性分级和肝纤维化评分明显降低(P<0.05)。结论:GHCCC对CCl4复合因素致肝纤维化大鼠具有一定的治疗作用。Objective : To investigate the effects of GHCCC in HF model rats induced by carbon tetrar - chloride com-pounded with other factors. Methods : One hundred SD rats excluding ten rats in blank control group were injected carbontetrar - chloride ( CC14 ) hypodermically and were fed with high fat diet and 10% ethyl alcohol every other day for eightweeks to establish HF models. Existing rats were divided into six groups randomly after four weeks:the normal controlgroup, the model control group, the positive control group ( 800mg·kg-1·d-1 ), the high, middle and low - dose groups( 800,400,200 mg·kg-1·d-1 ). After 8 - week treatment,the hyaluronic acid (HA) ,laminin (LN) ,type ]lI precolla-gen ( ⅢPC) and hydroxyptoline (Hyp) were assayed according to the manufacturer's instructions and the histopatholog-ical changes were observed by MASSON staining. Result : GHCCC groups' HA, LN and HI PC in serum and collagen fibersin hepatic fibrosis were significantly lower than those of the model group. The high does group Hyp level was lower thanthat of the model group and the hepatic pathological damage was relieved. The high and middle dose groups'fatty degener-ation of liver and liver fibrosis score were significantly reduced ( P 〈 0. 05 ). Conclusion : GHCCC was effective in treatmentof HF model rats induced by carbon tetrar - chloride compounded with other factors.
