摘 要:目的研究白芍总苷对金黄色葡萄球菌及大肠杆菌体外培养生物膜(biofilm,BF)形成及清除作用的影响,为中药白芍总苷开发利用及临床辅助治疗提供实验依据.方法:通过微孔板改良法构建金黄色葡萄球菌及大肠杆菌BF 体外模型,采用试管二倍稀释法测定白芍总苷最低抑菌浓度(MIC),研究其对两种细菌BF 形成的影响;同时通过检测对成熟BF 中活菌数的影响,评价白芍总苷对BF 的清除作用. 结果:白芍总苷对金黄色葡萄球菌及大肠杆菌的MIC 分别为4.48mg/ml 和9.38mg/ml.白芍总苷可在早期阶段干扰细菌BF 的形成;白芍总苷还可以抑制并有效清除成熟BF,其高浓度抗菌活性与青霉素/庆大霉素相比无显著性差异. 结论:白芍总苷在体外具有抑制细菌BF 形成的作用,并能通过破坏已形成的BF 起到杀灭相应细菌的作用,为临床辅助治疗耐药性感染,提供实验依据.ObjectiveTo study the inhibit function of vitro Staphylococcus aureus and Escherichia coli biofilm (biofilm, BF) by total glucosides of paeony, provide theexperimental basis of clinical application. Methods:Induced the BF in vitro. Minimal inhibitory concentrations (MICs) were measured by doubling dilution. Staphylococcus aureus andEscherichia coli were treated with different diluted total glucosides of paeony, the formation and growth of Staphylococcus aureus and Escherichia coli BF were tested bydetermination of the OD value and viable bacterial counts. Results:The MIC of Staphylococcus aureus and Escherichia coli by total glucosides of paeony are 4.48mg/ml and9.38mg/ml,respectively. Total glucosides of paeony could inhibit the formation of Staphylococcus aureus and Escherichia coli’s BF and they could destroy the mature BF effectively.Conclusion:Total glucosides of paeony can effevtively inhibit the formation of Staphylococcus aureus and Escherichia coli’s BF in vitro. It can also destroy the mature BF.
