摘 要:目的观察补肺通络疗法对内毒素小鼠肺纤维化的治疗作用及其对肺组织中结缔组织生长因子(CT-GF)、α-平滑肌肌动蛋白(α-SMA)表达的影响。方法内毒素腹腔注射法制备小鼠肺纤维化模型,随机分为模型组、理气组、活血组、滋阴组以及补肺通络全方组,分别给予等体积生理盐水或相应药物灌胃,每日1次。于第14天和第28天处死小鼠,苏木精一伊红染色观察肺组织形态学改变,Masson染色观察肺组织纤维化程度,免疫组化方法检测CTGF和α-SMA的表达。结果给药第14天,各组仍存在明显的炎症反应,活血组肺损伤评分高于模型组(P〈0.01),其他各组与模型组相比差异无统计学意义(P〉0.05)。全方组肺组织纤维化水平最高,理气组和滋阴组次之,模型组和活血组最弱。CTGF表达活血组最高,其他各组间差异无统计学意义(P〉0.05);α-SMA表达全方组最高,模型组最低,其他各组间差异无统计学意义(P〉0.05)。给药第28天,与模型组相比,全方组炎症恢复最快(P〈0.01),理气组(P〈0.01)和滋阴组(P〈0.05)次之,活血组差异无统计学意义(P〉0.05)。各组纤维化水平均较前有所增加,以全方组增幅最小,模型组和活血组增幅最大。各组CTGF表达水平均有所降低,以全方组最为显著,而模型组和活血组仍处于较高水平。各组α-SMA表达水平均下降,以全方组和理气组最为显著,模型组和活血组仍处于较高水平。结论补肺通络综合疗法通过促进炎症反应消退、减少促纤维化因子CTGF和α-SMA的表达而发挥抗纤维化作用。Objective To observe the therapeutic effect of invigorating lung and promoting the collaterals (ILPC) prescription on endotoxin -induced pulmonary fibrosis in mice and its effect on connective tissue growth factor (CTGF) and α - smooth muscle actin ( α - SMA) expression in the lung tissue. Methods Endotoxin was given to the mice intraperitoneally, 10 mg/kg. Then, the injected mice were divided randomly into five groups: control (C) group, regulating Qi (RQ) group, activating blood (AB) group, nourishing Yin (NY) group and ILPC group. Each group were given corresponding prescriptions or normal saline, orally and daily. Mice were then sacrificed on d14 and d28. H - E stain was performed to observe the morphologic change of the lung. Masson stain was used to observe the degree of pulmonary fibrosis. Immunohistochemical method was used to detect the expression of CTGF and α- SMA. Results In d14, inflamma- tory responses were still obvious in each group, but only the lung injury score for AB group was higher than the control group (P 〈0.01 ). For the degree of pulmonary fibrosis, ILPC group was the highest, then were the RQ group and NY group. The control group and AB group were the weakest. For CTGF expression, AB group was the highest and no statis- tically significant difference was found among other groups ( P 〉 0.05 ). For α - SMA expression, ILPC group was the highest and the control group was the lowest, no statistically significant difference was found among other groups (P 〉 0. 05). In d28, when compared with the control group, resolution of inflammatory response was more apparent in ILPC group (P 〈 0.01 ), then, in RQ group (P 〈 0. 01 ) and NY group (P 〈 0.05 ). No statistically significant difference was found between AB group and control group ( P 〉 0.05 ).
