摘 要:目的观察混合噬菌体模拟抗原用于华支睾吸虫病诊断的价值。方法用斑点免疫金银染色法(Dot—IGSS)检测、筛选出阳性的噬菌体单个克隆,将免疫活性强的克隆组合为不同的混合噬菌体抗原。分别用华支睾吸虫成虫抗原和混合噬菌体抗原检测采自华支睾吸虫病流行区居民血清中抗华支睾吸虫抗体。结果用免疫反应性最强的噬菌体克隆P1+P6+P9组合作为混合噬菌体抗原,以Dot—IGSS法检测680份流行区居民血清,混合噬菌体抗原检测和成虫抗原检测的抗体阳性率分别为17.20%和19.26%,差异无统计学意义。结论混合噬菌体模拟抗原与华支睾吸虫成虫抗原有相似的敏感性和特异性,并可在实验室成批制作生产,有助于提高华支睾吸虫病免疫诊断的效能。Objective To investigate the application of mix phage displaying mimotopes in diagnosing Clonorchiosis. Methods Positive phage clones were screened out using Dot - IGSS. Then the clones with high activities were combined into mix phage antigens. The mix phage antigen and Clonorchis sinensis antigen were adopted to detect the presence of anti - Clonorchis sinensis antibody in the serum collected from patients infected with Clonorchis sinensis. Results The phage clone P1 + P6 + P9) presented the highest immunoreactivity, which was then adopted as a mix phage antigen to detect the serum samples from 680 patients using Dot - IGSS. The antibody positive rate was 17.20% and 19.26% for the mix phage antigen and Clonorchis sinensis antigen, respectively, without statistical differences ( P 〉 0.05 ). Conclusion Mix phage displaying mimotopes share the same sensitivity and specificity with Clonorchis sinensis antigen, which can be produced on a large scale and consequently improve the diagnosis of Clonorchiosis.
