摘  要:目的检测黄芩苷治疗后鼠巨细胞病毒(MCMV)感染模型鼠脾细胞中TBX21、GATA-3蛋白的表达强度,进一步研究黄芩苷在调节Th1/Th2平衡中的作用。方法建立MCMV播散性感染模型,在MCMV接种后24h给予黄芩苷腹腔注射60mg/kg,1次/d,共14d,并设立安慰剂对照组和正常对照组。分别于治疗后3、7和14d取小鼠脾脏,提取脾组织蛋白,用Western blot法分别检测Th1和Th2细胞特异性转录因子TBX21和GATA-3表达强度。结果治疗后3d,黄芩苷治疗组TBX21蛋白表达水平明显高于正常对照组和安慰剂对照组(P<0.01)。治疗后7d和14d,黄芩苷治疗组TBX21蛋白表达水平明显高于正常对照组(P<0.01,P<0.05);而黄芩苷治疗组与安慰剂对照组比较,差异无统计学意义(P>0.05)。治疗后3d,黄芩苷治疗组GATA-3蛋白表达水平明显低于正常对照组(P<0.01);而与安慰剂对照组比较,差异无统计学意义(P>0.05)。治疗后7d和14d,GATA-3蛋白表达水平3组之间差异无统计学意义(P>0.05)。结论黄芩苷在MCMV急性感染早期可明显上调TBX21蛋白表达,进而促进Th1细胞免疫应答,有利于机体清除病毒,并对MCMV诱导的Th1/Th2细胞网络改变有一定调节作用。Objective To detect expression intensity of protein TBX21/GATA‐3 in murine cytomegalovirus(MCMV)‐in‐fected mouse splenic cells after baicalin treatment ,in order to study the role of baicalin in regulating Th1/Th2 balance.Methods The disseminated infection animal model was established as follows :BALB/c mice of 4 weeks old were intraperitoneally injected with the MCMV Smith strain ,which was propagated in the immunosuppressive mice to increase the virulence.Baicalin of 60 mg/kg was injected intraperitoneally 24 h after the inoculation of MCMV ,once a day and totally for 14 days.The placebo group and normal control group were established simultaneously. Protein was extracted from murine spleen tissue 3 ,7 and 14 days af‐ter the treatment. The protein expression of TBX21 and GATA‐3 specific for Th1 and Th2 cell was detected by Western blot.Results The expression of TBX21 protein in placebo group was a little lower than that of normal control on day 3 ,but ob‐viously higher on day 7 and 14.On day 3 ,the expression of TBX21 protein was significantly higher in baicalin‐treated group than in normal control and placebo group(both P〈0.01).TBX21 protein of baicalin‐treated group was significantly higher than that of normal control on 7th and 14th day after treatment(P〈0.01 ,P〈0.05) ,but it had no significant difference as compared with placebo group(P〉0.05). The expression of GATA‐3 protein of placebo group and baicalin‐treated group was significantly lower than that of normal control on day 3 after treatment ;however ,on day 7 and 14 ,both were similar to that of normal con‐trol ,and there was no significant difference among the three groups.Conclusion During the early period of acute MCMV infec‐tion ,baicalin can promote Th1 cell immunity by increasing the expression of TBX21 protein ,so as to clear the virus and modu‐late the change of Th1/Th2 cell net caused by MCMV.
