摘  要:利用大孔吸附树脂对独活(Heracleum hemsleyanum Diels)中的总香豆素类成分进行纯化。采用静态与动态吸附-解吸相结合的方法,以解吸量及解吸率为主要指标对工艺条件进行优化,确定了最佳纯化工艺条件。结果表明,采用LX-36型大孔吸附树脂纯化效果较好,其最佳工艺条件为上柱药液浓度相当于原药0.1 g/m L,上柱药液p H为2.5-5.5,吸附速率为5 BV/h,上样量相当于树脂量的50%,解吸液乙醇体积分数为95%,解吸速率为1 BV/h,解吸液用量为5 BV,经LX-36型大孔吸附树脂分离纯化后的独活干浸膏中总香豆素的含量由原来的8.42%升高到27.09%。表明LX-36型大孔吸附树脂适于独活总香豆素的初步纯化。The technology of purifying total coum arins(TC) in Heracleum hemsleyanum Diels with macroporous adsorption resin was optimized.Using static and dynamic state combined with desorption amount and desorption ratio method,the optimum purification conditions with macroporous adsorption resin were determined using adsorption and desorption rate of TC as evaluating indicators.The results showed that LX-36 macroporous adsorption resin had better purification effect than others.The TC content of the sample(equal to raw material),p H value of sample,adsorption rate,amount of sample,ethanol content,desorption rate,the volume of eluant were 0.1 g/m L,2.5 to 5.5,5 BV/h,50%(equal to the resin),95%,1 BV/h,and 5 BV,respectively.Under these conditions,the purity of TC varied from 8.42% to 27.09%.It is indicated that LX-36 macroporous adsorption resin was suitable for initial purification of TC in Heracleum hemsleyanum Diels.
