经穴注射骨髓间充质干细胞联合中药对糖尿病大鼠后肢缺血血流的影响

摘　　要：目的:探讨经穴注射骨髓间充质干细胞(BMSCs)联合中药对糖尿病大鼠后肢缺血血流的影响。方法:80只SD大鼠随机分为8组:正常假手术组、糖尿病(DM)假手术组、DM缺血组、中药组、局部注射组、局部+中药组、穴位注射组、穴位+中药组。注射选穴:三阴交、照海、环跳、阳陵泉、后三里;局部注射取缺血组织与正常组织交界处等距离选取5个点为注射部位。每只大鼠共注射1×107/m L BMSCs 100μL,每处注射20μL,其他对照组注射等量PBS缓冲液。应用Peri Scan PIM型激光多普勒血流成像仪,比较各组大鼠后肢不同时期血流指数(PUI)变化,以及分析中药与注射BMSCs的交互作用。结果:组内PUI比较,穴位注射组、穴位+中药组术后7d与术后2d比略有升高(P<0.05);DM缺血组、中药组、局部+中药组、穴位注射组、穴位+中药组术后14、21d大幅升高(P<0.05),且穴位+中药组PUI术后21d显著升高(P<0.01),差异均有统计学意义。术后7d、14d、21d血流指数差值与术后2d组间比较,穴位+中药组明显较正常假手术组、DM假手术组升高(P<0.05),差异均有统计学意义。中药与注射BMSCs的交互作用不明显。结论:经穴注射BMSCs联合中药或明显改善糖尿病大鼠后肢缺血血流情况,比其他方法对于促进后肢血流量效果好,其次是穴位注射组,然后是局部+中药组、局部注射组、中药组。Objective： Explore the effects of acupoint injection of bone marrow mesenchymal stem cells （BMSCs） transplantation combined Chinese Herb on blood flow of diabetic rats model with hind limb arterial ischemia. Methods： 80 SD rats were randomly divided into 8 groups： Normal control group （NC）, Diabetic rats control group （DC）, Diabetic rats ischemic group （DI）, Qi-tonifying and Blood-activating Chinese herb treatment group （QBT）, BMSCs Local injection group （LI）, BMSCs local injection combined Chinese herb group （LI＋QBT）, BMSCs acupoint injection group （AI） and BMSCs acupoint injection combined Chinese herb group （AI＋QBT）. Acupoint of ‘Sānyīnjiāo＇（三阴交SP6）, ’Hòusānl＇ （后三里ST 36）, ‘Zhàohǎi＇ （照海KI6）, ‘Huántiào＇（环跳GB30） and ‘Yámglíngquán＇ （阳陵泉GB34） were selected for acupoint injection, and equidistance selected 5 points at the junction of ischemic and normal tissues for local injection. Quantity： 100μL 1× 10^6cells/per rat, 20μL/per point, other control groups injected the same volume PBS buffer. The changes of hind limb blood flow （perfuse unit, PU） of each groups were consecutively detected by using laser Doppler flowmetery. The PU indexes （PUI） were compared between groups, and analysis the interaction of Qi-tonifying and Blood-activating Chinese herb treatment with injection BMSCs. Results： The comparison of PUI was slightly elevated in AI group and AI＋QBT group after 7d of postoperative compared to 2d postoperative, and sharply raised at DI, QBT, LI＋QBT, AI, AI＋ QBT group after 14d and 21d of postoperative （P〈0.05）, AI＋QBT raised significantly at 21d （P〈0.01）. Compared the PUI difference value of 7d, 14d, 21d postoperative with 2d＇s separately, AI＋QBT group raised obviously compared to NC and DC groups （P〈0.05）. The interaction between QBT and BMSCs injection was not obvious, Conclusion： BMSCs acupoint injection combined Chinese herb can obviously
