摘 要:目的:观察甘露消毒丹对H1N1流感病毒感染小鼠肺和血清中细胞因子干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)、白介素-2(IL-2)及白介素-6(IL-6)表达的影响,探讨其抗流感病毒感染的作用机制。方法:BALB/c小鼠分为4组:正常对照组和病毒模型组以0.9%氯化钠溶液灌胃,达菲对照组和甘露消毒丹组以各自水溶液灌胃。除正常对照组外,余3组以FM1-6-E2滴鼻造模。每组于造模后3、6、9d分别取肺和血液制备肺匀浆和血清,采用ABC-ELISA法测定各细胞因子变化。结果:各组与同期病毒模型组比较,除正常对照组第3天血清中IFN-γ、第9天血清中IFN-γ和IL-2,达菲对照组第3天肺和血清中IL-6、第9天血清中IFN-γ,甘露消毒丹组第3天肺中IL-2、血清中TNF-α、IL-2、IL-6外,余各组各时段细胞因子均有显著差异(P<0.01,P<0.05)。结论:甘露消毒丹可有效增强IFN-γ、IL-2和抑制TNF-α、IL-6的异常表达,可一定程度上正面调节机体的免疫功能。Objective: To observe the effects of Ganluxiaodudan on the expression of cytokines IFN- γ,TNF- α, IL-2 and IL-6 in the lung and serum of mice that infected with H1N1 influenza virus to explore its mechanism of anti influenza virus infection. Methods: BALB/c mice were divided into 4 groups: Mice stomachs were perfused with saline in normal control group and virus infected model group. Tamiflu group and Ganluxiaodudan group were respectively perfused stomach with their solution. The model mice were induced by FM1-6-E2 nose drops in the groups except the normal control group. At 3d, 6d, 9d after modiing every group were obtaind lung homogenate and blood serum from the lung and blood. Determination of the changes in these cytokines by ABC- ELISA. Results: Except the level of IFN-γ in blood serum at 3d and the level of IFN- γ and IL-2 in blood serum at 9d in normal control group, the level of IL-6 in lung and blood serum at 3d and the level of IFN- γ in blood serum at 9d in tamiflu group, the level of IL-2 in lung and the level of TNF- α, IL-2, IL-6 in blood serum at 3d in Ganluxiaodudan group, there have statistically significant differences of the level of cytokines at each time period in groups compared with virus infected model group (P〈0.01, P〈0.05). Conclusion: Ganluxiaodudan can enhance the expression of IF- γ, IL-2 effectively and depress the abnormal expression of TNF- α, IL-6. To a certain extent Ganluxiaodudan can also modulate immune function positively.
