摘 要:目的:探讨缺血性中风病血瘀证与蛋白质组学的相关性。方法:选择30例缺血性中风病患者,抽取血液标本,采用双向电泳和质谱技术对其蛋白质组进行分离和鉴定,利用数据挖掘的方法与血瘀证候相关联,分析得出差异表达蛋白质点。结果:缺血性中风病急性期血瘀证患者血浆和非血瘀证患者血浆相比,血浆2-DE凝胶图片中共发现了6个差异表达蛋白质,其中有5个蛋白质表达量上调,分别是结合珠蛋白、SP40/40、血管Rab-GAP/TBC-containing蛋白、纤维蛋白原γ链、γ-肌动蛋白;1个蛋白质表达量下调:TROVE域家族,成员2。结论:蛋白组学异常是出现相应中医证侯的一个重要危险因素。本研究结果中得出的差异表达蛋白,希望可以为今后研究缺血性脑卒中患者的生物学信息提供线索。Objective: To investigate the correlation between blood stasis syndrome and proteomics of ischemic stroke. Methods: Blood samples of 30 ischemic stroke patients were collected. The protein was separated and identified by using two- dimensional electrophoresis and mass spectrometry, and the differentially expressed proteins were analyzed by using the correlation between data mining method and blood stasis syndrome. Results: Six differentially expressed protein were discovered between the acute ischemic stroke patients with syndrome of blood stasis and acute ischemic stroke patients with non-syndrome of blood stasis by using 2-DE gel image of plasma. There were 5 protein expressions increased such as haptoglobin, SP40/40, Rab-GAP/TBC-containing, fibrinogen γ chain and γ-actin, and expression of one protein (TROVE domain family, member 2) decreased. Conclusion: Abnormal proteomics is an important risk factor of appearance of corresponding anomaly syndromes. The differential proteins obtained in this study might provide clues for the future research on biological information of patients with ischemic stroke.
