摘  要:目的:对脾虚证代谢综合征大鼠血清蛋白组学进行分析,并探讨脾虚证代谢综合征的微观信息,找寻生物学基础。方法:采用同位素标记技术,利用高效液相色谱仪及四级杆-静电场轨道阱质谱仪,对脾虚证代谢综合征大鼠进行串级质谱数据分析,对其血清蛋白质表达进行研究。结果:通过实验研究,共获得脾虚证代谢综合征大鼠模型50个差异蛋白,主要有免疫球蛋白Ig G、肉碱乙酰转移酶、羧酸酯酶等,其中免疫球蛋白Ig G、肉碱乙酰转移酶、羧酸酯酶等较空白对照组出现了上调,而谷胱甘肽S-转移酶、硒结合蛋白等出现了下调。结论:脾虚证代谢综合征存在体内免疫系统,糖类、脂类代谢紊乱,血清蛋白组学分析对探讨脾虚证代谢综合征生物学理论基础有重要意义。Objective: By analyzing the proteomics of rats with metabolic syndrome with spleen deficiency syndrome in serum and explorement of its microscopic information, to research its biological basis. Methods: The serial spectrometry data of rats with metabolic syndrome with spleen deficiency syndrome were obtained with the isotope labeling technique, high performance liquid chromatography and four bar-orbitrap mass spectrometer techniques. Results: There were 50 differential proteins in rats with metabolic syndrome with spleen deficiency syndrome, such as immunoglobulin G (IgG), carnitine acetyltransferase, carboxylesterase and so on. The IgG, carnitine acetyltransferase and carboxylesterase in spleen deficiency group were higher than those in the normal group, while the glutathione S-transferase, selenium binding protein were lower. Conclusion: There are disorders in immune system, carbohydrate and lipid metabolism in metabolic syndrome with spleen deficiency syndrome. The serum proteomic analysis is significant for exploring the biological theory of metabolic syndrome with spleen deficiency syndrome.
