摘  要:atp1基因在植物中是线粒体基因组编码,其产物ATP1是线粒体ATP合酶F1的α亚基,在小麦BNS的雄性不育系和它的转换系中差异表达。为了检测该基因的表达丰度,探讨与BNS不育性的联系,以BNS不育系和它的转换系的幼穗、花药和穗轴等组织为材料,利用实时荧光定量PCR方法,测定和比较该基因在不同组织中的表达水平,结果表明,该基因在各组织中的总表达量比内参甘油醛-3-磷酸脱氢酶基因表达量高3-4个数量级;在幼穗及各穗轴营养组织中表达量一致;花药与营养组织比较,在不育系四分体和单核期花药中表达均上调;在不育系中,该基因表达量在单核期花药中显著下调。这些结果说明,线粒体atp1基因在小麦中是一个高水平表达基因,在BNS营养组织中组成型表达,在花药中特异性上调表达,在不育系中表达受到抑制,表现显著下调,表明atp1基因的下调表达与BNS不育性有关。atp 1 is a mitochondrial genomic gene in plants, that encode a protein, ATP1, called α subunit of mitochondrial ATP synthase F1, and expresses differentially betweem BNS male sterile lines and its conversion line in wheat. In order to find the relationship between the gene expression and BNS infertility, the expression abundance of the gene was detected in the tissues of young panicles, anthers and ear axis of BNS male sterile line and its conversion line by the method of fluorescence real-time quantitative PCR. The re- sults showed that the expression amount of the gene in ear axis and young panicle tissues was at the same level, and up-regulated in anthers of tetrad and mononuclear stage in the fertile lines, but significantly down-regulated in mononuclear stage of the sterile line. The total expression amount of the gene was higher at the level of 3 to 4 ACt than that of the internal reference gene, glyceraldehyde-3-phosphate dehydrogenase. These results suggested that mitochondrial atp 1 gene in wheat BNS was a high level expression gene, and it expressed constitutive in vegetative tissues, and had the characteristics of specificity in anthers, and down-regulated expression in sterile line, indicating the genes of atp i and its expression amount was related to the infertility of BNS.
