摘 要:目的探讨知柏地黄汤对解脲支原体(ureaplasma urealyticum,UU)感染大鼠生精细胞线粒体琥珀酸脱氢酶(SDH)的影响。方法将40只SD雄性大鼠,随机分成正常组和造模组,采用经膀胱注射UU标准株悬液接种,建立UU感染动物模型,再将造模成功的大鼠随机分为模型组、阿奇霉素组(西药组)、知柏地黄汤组(中药组)。采用彩色精子动态检测系统检测精子运动参数;ELISA方法检测SDH的活性及含量;RT-PCR法检测SDH m RNA表达。结果与正常组比较,模型组大鼠精子活力、SDH活性和含量以及SDH m RNA表达均明显降低(P<0.01)。与模型组比较,中药组和西药组精子活力及SDH含量均提高(P<0.05,P<0.01),中药组SDH活性提高(P<0.01),西药组SDH m RNA表达提高(P<0.05)。中药组在提高精子运动参数方面效果优于西药组(P<0.05,P<0.01)。结论知柏地黄汤改善UU感染大鼠精子活力的效果优于阿奇霉素,可能通过升高SDH活性与含量来达到提高精子活力的作用。Objective This study was to explore the effects of Zhibai Dihuang Decoction on spermatogenic cell mitochondrial succinate dehydrogenase of rats infected by ureaplasma urealyticum (UU). Methods The 40 SD rats were randomly divided into normal control group and model group, their bladders were inoculated with UU suspension to establish the UU infection models. Then successful model rats were randomly divided into model group, the azithromycin group (western medicine group), and Zhibai Dihuang Decoction group (TCM group). Sperm motility parameters were detected by the color sperm dynamic detection system; The activity and content of succinate dehydrogenase (SDH) were tested by ELISA; The expressions of SDH mRNA were detected by RT-PCR method. Results The sperm motility, the activity and content of SDH and the expression of SDH mRNA in the model rats are significantly lower than the control group (P〈0.01). Comparing with the model group, the Rat sperm motility and the content of SDH in western medicine group and TCM group (P〈0.05, P〈0.01); the SDH in Zhibai Dihuang Decoction group was improved (P〈0.01), while SDH mRNA in azithromycin group was improved (P〈0.05). Curvilinear velocity and average velocity of Zhibai Dihuang Decoction group were better than azithromycin group (P〈0.05, P〈0.01). Conclusion The sperm motility in UU affection rats treated with Zhibai Dihuang Decoction was better than that with azithromycin. Zhibai Dihuang Decoction increase sperm motility maybe through increasing the content and activity of SDH.
