摘  要:目的:观察通心络对脑缺血模型大鼠脑组织病理学及胎盘生长因子(placental growth factor,PlGF)表达水平的影响,探讨其治疗脑梗死的作用机制。方法:以线栓法制作大鼠大脑中动脉缺血模型(MCAO),将SD大鼠随机分为假手术组、模型组、通心络组及丁苯酞组,分别在造模后1 d和3 d进行神经功能缺损评分,取脑和采集动脉血。采用HE染色方法观察脑组织的病理变化;免疫组织化学及酶联免疫(ELISA)的方法,分别检测脑组织和血清中PlGF的表达水平。结果:与模型组比较,通心络组能显著改善大鼠神经功能缺失症状,减轻大脑皮质及海马神经元的损伤,脑组织和血清PlGF表达均明显增强(P<0.05)。结论:通心络对急性脑梗死损伤具有保护作用,其作用机制可能与增加PlGF的表达有关。Objective:To observe effects of Tongxinluo( TXL)on the pathologic change and the expression of placental growth factor in rats with acute cerebral infarction,and to explore its mechanism of action. Methods:Using filament method to copy middle cerebral artery occlusion( MCAO)model of rats. The rats were randomly divided into 4 groups:the sham-operated group,the model group,TXL group and Butylphthalide group. After modeling for 1 day and 3 days,the neurological scores were taken to evaluated the rats,the morphologi-cal feature of neurons was observed by HE staining,the level of PlGF on the brain and serum were detected by immunohistochemistry and ELISA. Results:Compared with the model group,the neurological impairment of the TXL group were alleviated,cell injury of the hippocampus and cortex were relieved,brain and serum expression of PlGF were increased(P〈0. 05). Conclusion:TXL has protective effects on rat cerebral ischemia brain injured by MCAO,which is related to regulating PlGF expression.
