水蛭素对凝血酶造成的血管内皮细胞单层通透性增高的抑制作用

摘　　要：观察水蛭素对凝血酶造成的血管内皮细胞单层通透性增高的抑制作用。从新生牛主动脉分离获得单个内皮细胞 ,培养于微孔滤膜上直至形成致密单层。通过灌流Hanks液或含 5g·L-1 白蛋白的Hanks液后 ,测定经 1 0U/ml凝血酶溶液处理或经 1 0U/ml凝血酶溶液加 1 0U/ml水蛭素处理的液体滤过系数 (Kf)、液体滤过流量(Jv)和蛋白质渗透压反射系数(σ)。结果 :内皮单层经凝血酶处理后 ,Kf和Jv降低 ,σ升高 ;而水蛭素能够抑制因凝血酶造成的Kf和Jv降低 ,以及σ升高。提示 :凝血酶能够引起血管内皮单层通透性的增加 ,而水蛭素能够减轻凝血酶造成的内皮单层通透性的增加 。To observe the function of hirudin in inhibiting the monolayer permeability due to thrombase,single endothelial cell isolated from aorta of newborn calf is cultured in microporous filtering film till compact monolayer is formed.After irrigation with Hanks solution possibly containing 5g·L -1 album,the Kf,Jv and σ of solutions,which are handled by 10U/ml thrombase as well as possible 10U/ml hirudin,were measured.Results:In endothelial monolayer handled with thrombase,Kf and Jv reduced,while σ increased;but hirudin could fight off the function of thrombase.It is indicated that thrombase can increase the monolayer permeability,while hirudin can prevent the permeability increase;the mechanism remains to be further studied.
