摘  要:目的:探讨霉酚酸酯对糖尿病肾病大鼠肾脏Toll样受体-4(TLR4)及白细胞介素-18(IL-18)表达的影响。方法:雄性SD大鼠随机分为对照组(n=12)、模型组(n=11)、治疗组(n=11),后2组制作糖尿病肾病模型。治疗组每天给予霉酚酸酯15 mg/kg灌胃,余2组给予溶媒灌胃。于12周末采用HE染色、PAS染色观察肾脏病理改变,免疫组化、Western blot检测肾组织TLR4、IL-18蛋白的表达。结果:与对照组比较,模型组大鼠肾小球体积增大,基底膜增厚,炎症细胞浸润明显,而治疗组较模型组有所改善。3组大鼠肾组织TLR4、IL-18蛋白的表达差异有统计学意义,其中模型组大鼠肾组织TLR4、IL-18蛋白的表达均高于对照组(P<0.05),治疗组上述2指标较模型组下降(P<0.05)。结论:霉酚酸酯可能通过下调肾脏TLR4、IL-18蛋白的表达对糖尿病肾病大鼠的肾脏起保护作用。Aim:To investigate the effects of mycophenolate mofetil (MMF) on the expressions of TLR4 and IL-18 in kidneys of diabetic nephropathy rats .Methods:SD rats were allocated into 3 groups:normal control group(n=12),dia-betic nephropathy group(model group,n=11),and treatment group(n=11).The latter 2 groups were established diabetic nephropathy model .The treatment group was given MMF at 15 mg/kg per day ,and the other 2 groups were given the sol-vent.Twelve weeks later , kidney pathology was observed using HE staining and PAS staining .Immunocytochemistry and Western blot were used to detect the expressions of TLR 4 and IL-18.Results:HE staining and PAS staining of nephridial tissue showed that glomerular volume ,thickness of glomerular basement membrane and infiltration of inflammatory cells in the model group increased significantly compared with the normal control group .Improvements were observed in the treat-ment group .The differences in the expressions of TLR 4 and IL-18 in nephridial tissue among three groups were statistically significant.The expressions of TLR4 and IL-18 in nephridial tissue in model group were higher than those in normal control group(P〈0.05).Conclusion:The protective role of MMF may be associated with the decrease of TLR 4 and IL-18 in dia-betic nephropathy rats .
