摘 要:本文建立了一种基于纸芯片的乳酸脱氢酶(LDH)检测方法。实验优化了黄递酶(DIA)/碘硝基四唑紫(INT)显色体系检测LDH的条件,考察了方法的选择性,获得了较好的线性关系,并成功用于血清样品中LDH含量的测定。同时,结合DIA/INT和BCIP/NBT显色体系,通过构建一维、三维纸芯片,实现了LDH和碱性磷酸酶(ALP)的同时检测。两者同时检测时,颜色区分明显,相互之间干扰较小。纸芯片显色法操作简单、结果直观可见、效果良好。The microfluidic paper-based analytical devices(μPADs)are a new kind of analytical platform that is made from paper patterned into hydrophilic channels bounded by hydrophobic polymers or others.Considerable attention has been paid toμPADs because of their low cost,disposable,equipment-free,ease of use,potential to be used remotely,and ability to provide results in telemedicine.In this work,a lactate dehydrogense(LDH)sensor has been established.The factors of DIA/INT system for LDH detection has been investigated and the good selectivity,calibration curves of the sensor were got.The sensor has been successfully applied for the detection of LDH in human serum samples.Based on DIA/INT and BCIP/NBT colorimetric system,the simultaneous detection of lactate dehydrogenase and alkaline phosphatase(ALP)was applied on 1Dand 3DμPADs.This low-cost and portable analytical device has been proved to be useful for the point-of-care application with good sensitivity and reproducibility.
