摘  要:【目的】建立以高效液相色谱-蒸发光散射法(HPLC-ELSD)测定西瓜果实糖分类型和含量的方法,为精准鉴定西瓜种质资源的糖分类型和测定其含量及西瓜品质育种提供参考依据。【方法】采用HPLC-ELSD法,设置色谱条件采用Comosil Sugar-D柱(4.6 mm ID×250.0 mm),流动相为乙腈-水(体积比75∶25),流速1.0 mL/min,柱温30℃;ELSD参数:漂移管温度80℃,气体流量1.60 mL/min,分析西瓜果实中可溶性糖的类型和含量。【结果】在10 min内准确检测出西瓜果实中葡萄糖、果糖和蔗糖3种糖,葡萄糖、果糖和蔗糖在线性范围0.25~3.00 mg/mL内线性关系良好,相关系数r果糖=0.9943、r葡萄糖=0.9997、r蔗糖=0.9988。加样回收率98.57%~101.36%;西瓜果实中果糖、葡萄糖和蔗糖含量分别为45.88、16.62和34.03 mg/g。【结论】HPLC-ELSD法具有操作简便、准确、重复性好等优点,可快速准确测定西瓜果实中糖的类型及其含量。【Objective 】The components and contents of water-soluble sugar in watermelon fruit were studied using HPLC- ELSD to provide references for accurate identification of watermelon germplasms and quality watermelon breeding. 【Method 】The components and contents of water-soluble sugars in watermelon fruit were analyzed by HPLCELSD. The working condition of HPLC was Comosil Sugar-D column(4.6 mm ID ×250.0 mm),acetonitrile and water with a volume ratio of 75 ∶25 as mobile phase, flow rate of 1.0 mL/min and column temperature of 30 ℃. ELSD parameters were detector drift tube temperature of 80℃ and gas flow rate of 1.60 mL/min. 【Result 】The contents of glucose,fructose and sucrose could be precisely measured in 10 minutes. The linear relation of fructose(r =0.9943),glucose(r=0.9997) and sucrose(r=0.9988) was good in the range of 0.25-3.00 mg. The recovery of sample ranged from 98.57% to 101.36%. The contents of fructose, glucose and sucrose in watermelon fruit were 45.88,16.62 and34.03 mg / g, respectively. 【 Conclusion 】 The method of HPLC- ELSD possesses advantages of easy operation, high accuracy and good reproducibility, and it can be applied as a quick measurement of the contents of glucose, fructose and sucrose in watermelon fruit.
