高效液相色谱法测定甘草内生真菌产芦丁条件优化的研究

摘　　要：通过优化利用高效液相色谱法(HPLC)测定甘草内生真菌产芦丁的方法条件,对从甘草中分离得到的产芦丁内生真菌进行研究,为后期利用HPLC法筛选产活性物质内生真菌的研究奠定基础。对产芦丁甘草内生真菌采用高效液相色谱法(HPLC)进行筛选,并通过条件优化找到检测产芦丁内生真菌的最优HPLC方法,找到HPLC最优条件组合筛选产芦丁的甘草内生真菌。HPLC最优条件组合为:流动相A为乙腈、流动相B为0.05%磷酸水溶液、柱温T=50℃、流动相比例有机相A(乙腈):流动相B(0.05%磷酸)=0.20∶0.80、波长λ=360nm、进样体积V=20μL。The preseant study opyomized the condition for determing rutin produced by endophytic fun‐gus in glycyrrhiza by high performance liquid chromatography （HPLC） and further researched the rutin fungus separated from glycyrrhiza to prepare for screening active - bio - mass - producing fungus by HPLC .Through selection ,optimization and comparioson ,the optimal condition for using HPLC were set‐ting Acetonitrile as mobile phase A ,0 .05% phosphoric acid as B at a ratio of 2/8 ;column temperature to be 50 degrees celcius ,wavelength 360nm ,and injecion volume 20μl .
