骨龙胶囊治疗类风湿性关节炎大鼠的作用机制研究

摘　　要：目的探讨骨龙胶囊对类风湿性关节炎大鼠血清白介素1β(IL-1β)和肿瘤坏死因子α(TNF-α)含量的影响,阐释其作用机制。方法运用Ⅱ型胶原加完全弗氏佐剂混合物皮内注射建立RA大鼠模型,用酶联免疫分析法检测大鼠血清IL-1β和TNF-α的含量。结果与空白组比较,模型组各项指标含量均明显增高,差异具有统计学意义(P<0.05);与模型组比较,骨龙胶囊组及甲氨蝶呤组各项指标含量均有不同程度下降,差异有统计学意义(P<0.05);与甲氨蝶呤组比较,骨龙胶囊组各指标含量下降,差异有统计学意义(P<0.05)。结论骨龙胶囊能有效抑制类风湿性关节炎大鼠血清IL-1β和TNF-α炎性细胞因子的活性,对类风湿性关节炎有良好的治疗作用。Objective To investigate the effect of Gulong capsule on serum IL - 1β and TNF - α level of rheumatoid arthritis rats as well as its action mechanism. Methods An experimental type Ⅱ collagen - induced arthritic rats was established. And serum IL - 1β and TNF - α levels were detected in experimental rats by enzyme - linked immunosorbent assay （ELISA）. Results Compared with blank group, the levels of serum IL - 1β and TNF - α of rats in model group were significantly higher, there had significant difference（ P 〈 0.05 ） ; compared with model group, the levels of serum IL - 1 β and TNF - α of rats in Gulong capsule group and Methotrexate group were decreased markedly, there had significant difference （ P 〈 0.05 ） ; compared with Methotrexate group, the levels of serum IL - 1β and TNF - α of rats in Gulong capsule group decreased, there had significant difference （P 〈 0. 05 ）. Conclusion The Gulong capsule could effectively inhibit the activity of serum IL - 1β and TNF - α in rheumatoid arthritis rats and has a good therapeutic effect on rheumatoid arthritis.
