Studies on m TOR Signaling Pathway on NAFLD Liver Tissue of SD Rat by the Way of Intragastric Administration on Resveratrol

摘  要:目的探讨改善非酒精性脂肪肝病(NAFLD)大鼠肝组织m TOR信号通路机制。方法建造NAFLD动物模型后,分别采用Western Blot测试肝组织m TOR、S6K1、PGC-1α等基因蛋白含量和Real-Time PCR相对定量分析测试肝组织m TOR、S6K1、PGC-1α等mRNA。结果肝组织m TOR、S6K蛋白条带灰度相对值,NSRE组同NS组比较非常显著性降低(P<0.01)、同NS组比较显著性降低(P<0.05),m TOR、S6K mRNA Real-Time PCR表达量,NSRE组同NS组比较均非常显著性降低(P<0.01)。肝组织Mfn2、NRF和PGC蛋白条带灰度相对值,NFRE组同NF组比较均非常显著性升高(P<0.01),Mfn2、NRF和PGC mRNA Real-Time PCR表达量,NSRE组同NS组比较,NFRE组同NF组比较非常显著性升高(P<0.01)。结论白藜芦醇联合游泳运动通过降低肝组织m TOR-S6K活性,使得肝组织线粒体活性标志物PGC、NRF和Mfn2增多,从而实现延缓NAFLD的发生与发展。Objective To explore the mTOR signaling pathway on liver tissue of SD rat model of dietary - induced NAFLD by the way of intragastrie administration on resveratrol. Methods After the construction of animal models of NAFLD, giving 8 - week swimming exercise on the SD rat model of dietary - induced NAFLD by the way of intragastric administration on resveratrol. SD rats were killed after 8 weeks , testing liver tissue mTOR,S6K1 ,PGC - 1α,Mfn2,NRF and theirs mRNA Real - Time PCR of each group SD rat, Results NSRE group of mTOR ,S6K and theirs mRNA Real -Time PCR expression in liver tissue compared with group NS, there was a very significant decrease( P 〈 0. 01 ). Therefore, NFRE group of PGC -1α,Mfn2 ,NRF and theirs mRNA Real- Time PCR expression in liver tissue compared with group NF, there was a very significant increase(P 〈 0. 01 ). Conclusion SD rats significantly delay of non - alcoholic fatty liver in effect by decreasing the activity of mTOR- S6K and increasing the activity of PGC -1α,Mfn2 and NRF.
