摘 要:目的研制一种简便、实用的撞击型脊髓损伤制模装置。方法取成年SD大鼠,随机分为3个模型组(10 g重量撞击头分别自2、3、4 cm高度撞击脊髓)和1个假手术组,每组25只。利用自制撞击器建立大鼠脊髓损伤模型。各组大鼠于术前1 d,术后3、7、14、28 d各个时间点随机选取5只进行斜板实验及BBB评分,观察大鼠后肢运动功能情况,并对术后7 d各组大鼠受损脊髓组织进行HE染色观察。结果假手术组各观察时间点的后肢运动能力均无统计学意义(P>0.05)。2 cm打击组行为学评价结果随时间延长呈逐步恢复趋势(P<0.05);3 cm和4 cm打击组术后7 d,行为学评价结果较之3d有所升高(P<0.05),而14、28 d的斜板实验值不再升高(P>0.05),两组28 d的BBB评分也仅回升至5分左右。HE染色结果显示随撞击高度增加脊髓受损程度加重。结论本装置从3 cm及以上高度下落对脊髓进行撞击可造成完全性脊髓损伤,而2 cm下落撞击脊髓所造成的损伤可随时间的推移逐渐恢复。Objective To develop an economical and practical impact- type spinal cord injury device and provide a reliable animal model for spinal cord injury study. Methods A new- type of spinal cord impactor was designed and hand- made to produce rat spinal cord injuries. Adult SD rats were randomly divided into 3 experimental groups( 10 g weight hitting spinal cord by hand- free falling from 2 cm,3 cm,and 4 cm distance) and sham operation group( control group) with 25 rats in each group. Five animals from each group were selected to perform the tilt table test and BBB scoring to observe the function damage of rat hind legs at 3,7,14 and 28 d after surgery and / or spinal cord injury. Rat damaged spinal cord tissues were observed by HE staining 7 d after operation. Results No significant difference of the tilt table test evaluation and BBB scoring among different time points in the control group was shown( P〈0. 05). The BBB scoring in 2 cm- impact group gradually recovered along with the time( P〉0. 05). The BBB scoring was more significantly improved in 3 cm- and 4 cm- impact groups 7 d after injury than that 3 d after operation. However,there was no significant difference of the tilt table test between 14 and 28 d after injury in 3 cm- and 4 cm- impact groups with the BBB scoring 28 after injury in these two groups arriving at 5. HE staining showed that the degree of spinal cord injury was aggravated with the increased impact height. Conclusion This impactor could produce repeated and reliable spinal cord injury. Specifically,the 10 g weight head falling from 3 cm or above causes complete spinal cord injury with no significant recovery. Damage caused by the 2 cm falling distance could gradually recover with time.
