摘  要:水稻粒型是与其产量直接相关的重要性状。在前期研究证实来源于特大粒水稻TD70和籼稻品种Kasalath的重组自交系(RIL)群体中检测到已克隆的粒长GS3和q GL3基因的基础上,通过TD70和Kasalath的GS3、q GL3基因序列差异分析,设计功能标记,检测240个RILs和不同粒长的6个粳稻、9个籼稻中GS3、q GL3基因类型及其效应。结果显示:TD70和Kasalath中的GS3基因和q GL3基因分别在第2外显子上的编码区+165位置和第10外显子上的编码区+1 092位置存在一个由碱基C到碱基A的单核苷酸替换,据此设计了四引物扩增受阻突变体系(Tetra-primer ARMS-PCR)标记,TD70的GS3和q GL3基因分别扩增为270 bp和145 bp条带及448 bp和288bp条带,Kasalath的GS3、q GL3基因分别扩增为270 bp和175 bp条带及448 bp和216 bp条带;240个RILs中含有GS3-T和q GL3-T基因的株系61个,含GS3-T和q GL3-K的株系48个,含GS3-K和q GL3-T的株系17个;不同粒型基因组合的粒长存在显著差异,表现为GS3-T+q GL3-T>GS3-T+q GL3-K或GS3-K+q GL3-T>GS3-K+q GL3-K;长粒型的1个粳稻和5个籼稻品种的GS3基因表现为TD70带型,短粒型的5个粳稻和4个籼稻品种的GS3基因表现为Kasalath带型;所有水稻品种的q GL3基因均表现为Kasalath带型。表明开发的GS3和q GL3基因功能标记是有效的,可用于水稻分子标记辅助育种。Grain shape is directly related to rice production.The cloned grain length genes GS3 and q GL3 in rice were detected in the recombinant inbred lines(RILs) derived from a cross between the extra-large grain japonica rice TD70 and the small grain indica rice Kasalath during early studies.In this study the functional markers were designed based on the sequence differences of GS3 and q GL3 between TD70 and Kasalath,and were used to test their gene types and effects in 240 RILs and some rice varieties including six japonica and nine indica.A single nucleotide transition from C to A was presented both in the second exon of GS3 and in the tenth exon of q GL3,and tetra-primer ARMS-PCR markers were designed according to this mutation,respectively.As a result,TD70 showed 270-bp and 145-bp fragments for GS3 gene,and 448-bp and 288-bp fragments for q GL3 gene,while Kasalath showed 270-bp and 175-bp fragments for GS3 gene,and 448-bp and 216-bp fragments for q GL3 gene.Among the 240 RILs,61 lines had GS3^-Tand q GL3^-Tgenes,48 lines had GS3^-Tand q GL3^-Kgenes,and 17 lines had GS3^-Kand q GL3^-Tgenes.Significant phenotypic differences existed between gene types,and the average phenotype values of grain length showed GS3^-T+q GL3^-TGS3^-T+q GL3^-Kor GS3^-K+q GL3^-T GS3^-K+ q GL3^-K.One japonica variety and five indica varieties with long grain showed GS3^-T type,while five japonica varieties and four indica varieties with short grain showed GS3^-Ktype.All these varieties showed the same type as Kasalath for q GL3 gene.The design of functional markers of GS3 and q GL3 was effective to detect 240 RILs and 15 rice varieties,and the markers could be used for rice molecular marker assisted breeding.
