脑健胶囊对局灶性脑缺血大鼠脑组织病理形态及血液流变学指标的影响

摘　　要：目的观察脑健胶囊对局灶性脑缺血大鼠脑组织细胞形态及血液流变学指标的影响。方法将大鼠随机分为假手术组、模型组、补阳还五汤组、脑健胶囊组,每组25只,采用大脑中动脉栓塞法(MCAO)建立永久性局灶性脑缺血模型,于手术后1 d、3 d、7 d、14 d、28 d处死,给予不同处理,采用HE染色法观察脑组织细胞形态、取动脉血进行血液流变学检测。结果 (1)模型组大鼠脑组织海马神经元细胞与假手术组比较出现典型缺血损伤表现,脑健胶囊组脑缺血组织与模型组比较自术后3 d开始恢复,且其起效时间优于补阳还五汤组。(2)模型组大鼠自术后1 d开始血液流变学中血浆黏度、全血高切相对指数、全血低切相对指数、卡松黏度即较假手术组明显升高(P<0.05或P<0.01),脑健胶囊组大鼠上述血液流变学指标在各时间点均较模型组降低(P<0.05或P<0.01),且其3 d的血浆黏度和28 d的全血高切相对指数水平改善均优于补阳还五汤组(P<0.05)。结论促进脑缺血组织细胞形态恢复,改善血液流变学特性,可能是脑健胶囊发挥脑保护作用的重要途径之一。Objective：To investigate the effect of Naojian capsules on the brain cells morphological distribution and hemorheology in rats after focal cerebral ischemia.Methods：The rats were randomly divided into sham operation group,model group,BYHWD group and Naojian capsules group,25 rats in each group.Establish a permanent focal cerebral ischemia model with the method of middle cerebral artery occlusion （MCAO）.On the 1 st,3 rd,7 th,14 th,28 th day after surgery,the rats were executed.Use HE staining to observe the rats＇ brain cells morphological distribution,and arterial blood for hemorheology detection.Results：（1）Compared with the sham group,the hippocampal neurons of the model group rats brain appeared typical ischemia tissue damage.Compared with the model group,the ischemia brain tissue of Naojian capsule group began to recover on 3rd day after surgery,which worked sooner than BYHWD group.（2）Compared with the sham group（P ＜ 0.05 or P＜0.01）,the plasma viscosity,whole blood high-shearing relative index,whole blood low-shearing relative index and Casson viscosity in hemorheology of the model group rats were all significantly increased on 1st day after surgery.Compared with the model group,the indicates above of hermorheology of the Naojian capsule could be reduced in each period （P＜0.05 or P＜0.01）.And the improving level of 3d Naojian capsule group of plasma viscosity and 14d Naojian capsule group of whole blood high-shearing relative index is better than BYHWD group （P ＜ 0.05）.Conclusion：Promoting the morphology distribution of ischemia brain tissue and improving the features of hemorheology may be an important way of protecting brain of Naojian capsule.
