溺毒清合剂对5/6肾切除大鼠TGF-β和Ⅳ胶原的影响

摘　　要：目的探讨中药经验方溺毒清合剂延缓慢性肾衰进展的作用机理。方法采用大鼠5/6肾切除造模方法复制慢性肾衰竭模型,对模型大鼠灌饲不同剂量的溺毒清合剂或厄贝沙坦,实验结束后查各组大鼠肾功能、尿蛋白排泄率,同时免疫组化法观察肾组织TGF-β和Ⅳ胶原蛋白的表达。结果溺毒清合剂和厄贝沙坦均可显著降低模型鼠的尿总蛋白和白蛋白排泄率,改善肾功能。免疫组化显示溺毒清合剂能降低肾组织中Ⅳ型胶原、TGF-β表达。结论中药溺毒清合剂可通过降低肾脏的尿蛋白排泄,抑制Ⅳ型胶原、TGF-β的表达,抵制肾间质组织的纤维化,从而延缓慢性肾衰竭进展。Objective：To study the mechanism of the traditional Chinese medicine named Niduqing mixture which can delay the process of chronic renal failure.Methods：The rat with 5/6 nephrectomy were used to set up the model of chronic renal failure,and the rats were randomly divided into two groups,given Niduqing mixture or irbesartan separately.Renal function and urinary albumin excretion were checked separately at the end of the experiment and at the same time,the TGF-β and the type Ⅳ collagens in renal tissue were observed with the method of immunohistochemical.Result：All of the Niduqing mixture and irbesartan can reduce urinary albumin excretion obviously in the rat model,and improve renal function.The Niduqing mixture can reduce the TGF-β and the type Ⅳ collagens in renal tissue by means of immunohistochemical analysis.Conclusion：The traditional Chinese medicine named Niduqing mixture can reduce urinary albumin excretion,suppress the TGF-βand the type Ⅳ collagens description,intervene renal interstitial fibrosis and then delay the process of chronic renal failure.
