摘 要:本试验旨在研究天冬氨酸(ASP)对脂多糖(LPS)刺激仔猪结肠黏膜结构和屏障功能的影响。试验选用24头仔猪,4个处理组分别为对照组、LPS组、0.5%ASP组、1.0%ASP组。试验第19天,分别给LPS组、0.5%ASP组、1.0%ASP组试验猪注射100μg/kg体重的LPS,对照组注射等量的生理盐水。注射后24 h屠宰,取结肠样品待测。结果表明:对照组仔猪结肠形态结构完整;注射LPS后,结肠上皮部分脱落;ASP组轻度脱落,与LPS组相比,0.5%ASP组结肠隐窝深度显著升高(P<0.05);0.5%和1.0%ASP可显著缓解LPS刺激导致的结肠杯状细胞数目的增多(P<0.05);0.5%ASP具有提高结肠中性粒细胞密度的趋势(P<0.10)。0.5%和1.0%ASP可显著缓解LPS刺激导致的结肠和血浆二胺氧化酶活性的降低(P<0.05)。以上研究结果表明,日粮添加ASP可有效阻止LPS对结肠结构和屏障功能的损伤。The study was conducted to evaluate the effects of aspartate (ASP) supplementation on colonic morphology and mucosal barrier function in piglets with lipopolysaccharide (LPS) challenge. Twenty-four piglets were randomly assigned into 4 groups: control; LPS; 0.5% ASP; 1.0% ASP. The trial period was 19 days. On day 19, the piglets in the LPS group, 0.5% ASP group and 1.0% ASP group were injected with 100 μg/kg BW LPS, and the piglets in the control group were injected with the same dose of sterile saline. After 24 h saline or LPS injection, all pigs were sacrificed to obtain colonic samples for analysis. The results showed that: the colonic morphology from piglets in the control group was intact. Colonic epithelium falled off in LPS group; Colonic epithelium in ASP group falled off to less degree. Compared with the LPS group, colonic crypt depth of 0.5%ASP was significantly increased (P〈0.05). 0.5% or 1.0% ASP alleviated the elevation of goblet cell number induced by LPS challenge (P〈0.05). 0.5% ASP tended to increase the density of colon neutrophils (P〈0.10). 0.5% or 1.0% ASP alleviated the decrease of DAO activity induced by LPS challenge (P〈0.05). Theses results indicate that ASP supplementation can prevent injury of colonic morphology and barrier function induced by LPS challenge.
