摘 要:目的探讨亚慢性氯化镧(La Cl3)暴露对子代大鼠海马组织核转录因子-κB(NF-κB)表达的影响。方法健康成年雌性Wistar大鼠24只,随机分为对照组及0.25%、0.5%、1.0%La Cl3组,染镧组仔鼠自雌鼠受孕至断乳后1个月通过自由饮水摄入镧;利用免疫组化法检测海马p-IKKα/β的分布和表达;应用Western blot法检测海马组织p-IκBα和p-NF-κBP65的表达。结果 0.25%、0.5%、1.0%La Cl3组仔鼠海马CA1区p-IKKα/β表达分别为(54.72±10.04)、(40.73±4.90)、(32.99±5.42),CA3区表达分别为(64.80±8.27)、(47.78±4.92)、(36.17±7.71),DG区表达分别为(303.67±34.78)、(166.83±29.8)、(87.01±17.40);CA1、CA3、DG区各染镧组仔鼠海马3区的p-IKKα/β表达与对照组比较,除DG区0.25%La Cl3组外其他各染镧组仔鼠海马p-IKKα/β表达均低于对照组(P<0.05);0.25%、0.5%、1.0%La Cl3组仔鼠海马p-IκBα和p-NF-κBp65蛋白表达IDV值分别为(0.58±0.06)和(0.46±0.06)、(0.49±0.07)和(0.36±0.08)、(0.36±0.09)和(0.25±0.08),均低于对照组的(0.68±0.05)和(0.56±0.08)(P<0.05),仔鼠海马p-IκBα和p-NF-κBp65蛋白表达均与La Cl3染毒剂量呈负相关(r=-0.994、-0.978,P<0.05)。结论亚慢性La C13染毒可引起子代大鼠海马细胞NF-κB活性降低,可能是镧引起学习记忆能力下降的机制之一。Objective To investigate the effect of lanthanum exposure on nuclear transcription factor-κB (NF-κB) expression in the hippocampi of offspring rats. Methods Female Wistar rats were obtained from the Center for Experi- mental Animals of China Medical University. The pregnant rats were exposed to lanthanum chloride(LaC13 ) (99. 9% ) in distilled drinking water at one of the four doses:0% ,0. 25% ,0. 5% ,and 1.0%. The offspring rats were exposed to LaC13 in utero and while nursing,and then they received 0,0. 25,0. 50 and 1.00% LaC13 orally in distilled drinking water for 1 month. Immunohistochemical assay was used to detect phosphorylated p-IKKα/β expression and distribution. The hipp- ocampi of offspring rats from the four groups were removed to evaluate p-IκBα and p-NF-KBP65 expression. Results Compared with those in the controls,the levels of p-IKKα/β expression in 0. 25% ,0. 5% and 1.0% LaC13 groups were 54. 72 ± 10. 04,40. 73 ±4. 90,and 32. 99 ±5.42 in hippocampal CA1 regionin,64. 80 ±8.27,47. 78 ±4. 92,and 36. 17 ± 7. 71 in hippocampal CA3 regionin, and 303.67 ± 34. 78,166. 83 ± 29. 8, and 87.01 ± 17.40 in hippocampal DG re- gionin. The levels of p-IKKα/β expression in LaC13 groups reduced significantly except for DG region of 0. 25% LaC13 group ( P 〈 0. 05 ). Intergrated density values ( IDVs ) of p-IκBα and p-NF-KBp65 protein expression in 0. 25 % ,0. 50% and 1.0% LaC13 groups were 0. 58 ±0. 06 and 0. 46 ±0. 06,0.49 ±0. 07 and 0. 36 ±0. 08,and 0. 36 +0. 09 and 0. 25 ± 0. 08 ,which were significantly lower than those in the control group(0. 68 ±0. 05 and 0. 56 ±0. 08) (P 〈0. 05). The ex- pressions of p-IκBα and p-NF-KBp65 in the hippocampi were negatively correlated to the exposure dose of LaC13 ( r = - 0. 994, - 0. 978 ; P 〈 0.05 ). Conclusion Subchronic LaC13 exposure lead to reduced NF-KB activity, and it may be one of the reasons of lower learning and memory ability resulted from lanthanum exposure.
