摘  要:目的:观察鹿茸对大鼠缺血下肢CD34+、FLK-1+阳性细胞比例的影响。方法:将54只SD大鼠随机分为3组:鹿茸组18只、模型组18只、假手术组18只。鹿茸组和模型组均采用左下肢股动脉结扎及分支剔除术,造成大鼠下肢缺血模型,假手术组仅暴露股动脉但不结扎股动脉及其分支。鹿茸组术后按0.571 g·kg-1·d-1剂量灌服,连续14 d。检测各组术前1d、术后第1、3、7、14天5个时间点大鼠外周血酶肌酸激酶(CK)、谷草转氨酶(AST)、乳酸脱氢酶(LDH)水平变化及双荧光阳性细胞(CD34+、FLK-1+)比例。结果:术后模型组外周血酶学指标CK、AST、LDH水平较假手术组均显著上升,术后第7、14天鹿茸组各项酶学指标水平较模型组及术后第1、3天均显著下降(P<0.05)。术后鹿茸组和模型组CD34+、FLK-1+双荧光阳性细胞比例较假手术组均显著升高(P<0.05),且鹿茸组升高较模型组更为显著(P<0.05或P<0.01)。结论:鹿茸可增加大鼠缺血下肢CD34+、FLK-1+阳性细胞的比例,改善下肢缺血状态,促进血管新生,其作用可能与其能增加外周血血管内皮祖细胞(EPCs)数量有关。Objective: To observe the Pilose antler on ischemic lower extremity CD34+ and the impact on FLK-1+ percentage of positive cells. Methods : 54 SD rats were randomly divided into three groups, Pilose Antler group, model group, the control group, with 18 rats in each group. Pilose antler group and model group adopted the left leg branch of femoral artery ligation and stripping out, causing lower limb ischemia rat model and controlled only exposed femoral artery but not ligation femoral artery and its branches. Postoperative Pilose antler set to 0.571 g ~ kg-1 ~ d-1 dose, administered orally for 14 consecutive days. Respectively before 1 day and 1,3,7,14 days after treatment, we detected the CK, AST, LDH levels and CD34+ and FLK-1~ proportion. Results: CK, AST, LDH levels of the three groups were significantly increased than those of control group. 7,14 days after operation and postoperative days 1,3, pilose antler group's enzymology indexes level compared with that of model group were significantly decreased ( P〈0.05 ). Pilose antler group and model group's CD34+ and FLK-1+ double fluorescence positive cells rates were significantly higher in the control group (P〈0.05), and deer antler group increases more significantly than those in the model group ( P〈0.01 ). Conclusion : Pilose antler can increase the rat ischemic lower limbsCD34+ and FLK-1+ proportion of positive cells, improve the state of lower limb ischemia and promote angiogenesis. Its effect and possible can increase the number of EPCs.
