摘 要:[目的]研究冷热护理刺激对人宫颈癌 H eLa细胞克隆形成能力的影响及机制,以期探索冷热护理的新作用机制,促进临床护理水平的提高。[方法]HeLa细胞按常规用DMEM 高糖培养基培养;按6孔板每孔100个~300个细胞的浓度接种并在不同培养温度情况下进行细胞克隆试验;提取在不同温度环境下生长的同批次 HeLa细胞的总 RNA,在逆转录反应后获得 cDNA,以此为模板,实施荧光定量分析不同温度环境下 pan QKI mRNA的表达水平。[结果]较高的环境温度能增强 HeLa细胞的克隆形成能力,抑制 pan QKI mRNA表达;但较低的环境温度则抑制了该细胞的克隆形成,同时,pan QKI mRNA表达水平显著增高。[结论]温度干预能对人宫颈癌 HeLa细胞的克隆能力产生影响,pan QKI 在不同温度环境中的表达变化可能是导致 HeLa细胞克隆形成能力差别的重要原因,温度干预的护理效用和机制值得深入探究。Objective:To probe into the influence of hot and cold care stimulus on forming ability of human cervi-cal cancer HeLa cells clone and its mechanism,so as to explore the new action mechanism of hot and cold care promote the improvement of clinical nursing level.Methods:HeLa cells were cultured by routine use of DMEM high glucose medium.According the concentration of 100~300 cells per hole,they were inoculated in 6 hole plate,cell cloning experiments were carried out in different culture temperature circumstances;the total RNA of the same batch of HeLa cells was extracted which grew in different temperatures,the cDNA obtained in the re-verse transcription reaction was taken as a template,the fluorescent quantitative analysis was implemented for expression level of pan QKI mRNA under different temperature environment.Results:The high ambient tem-perature can enhance the colony formation ability of HeLa cells and inhibit the pan QKI mRNA expression;but the lower ambient temperature inhibited the cell clone formation,at the same time,the expression level of mRNA was significantly increased in pan QKI.Conclusion:Temperature intervention can affect cloning ability of human cervical cancer HeLa cells.The changes of pan QKI expression in different temperature environ-ments may be an important reason of differences of HeLa cell clone forming ability.The efficacy and mechanism of temperature intervention are worth for in depth inquiry.
