摘 要:目的建立金钗石斛HPLC的特征图谱,为其质量评价提供参考。方法采用ZORBAX SB-Aq色谱柱;流动相乙腈-0.2%(体积分数)甲酸,梯度洗脱,流速1.0 m L·min-1;柱温30℃;检测波长270 nm。结果分别建立了不同产地金钗石斛的HPLC特征图谱,一类是以贵州赤水产金钗石斛为代表的HPLC特征图谱,标记出22个共有峰,相似度为0.979~0.998;一类是以云南产金钗石斛为代表的HPLC特征图谱,标记出27个共有峰,相似度为0.902~0.980。结论金钗石斛的HPLC特征图谱主要分为2类,贵州赤水产为一类,云南产为一类,广西产的两类都有。本法稳定可靠、重复性好,可用于不同产地金钗石斛的鉴别和质量控制。Objective To establish HPLC characteristic spectrum and provide reference for the quality evaluation of Dendrobium nobile Lindl.. Methods Chromatographic separation was performed on ZORBAX SB-Aq column at 30 ℃ eluted with acetonitrile and 0. 2% methanoic acid in a gradient elution at a flow rate of 1. 0 m L·min-1. The detection wavelength was set at 270 nm. Results The HPLC characteristic spectrum of D. nobile from different regions were constructed. One represented the characteristic spectrum of D.nobile from Chishui,Guizhou province with 22 common specific peaks,and the similarities were 0. 979- 0. 998.The other represented the characteristic spectrum of D. nobile from Yunnan province with 27 common specific chromatographic peaks,and the similarities were 0. 902- 0. 980. Conclusion HPLC characteristic spectrum of D.nobile could be divided into two major classes,one was from Chishui,Guizhou Province,and the other was from Yunnan Province. Samples from Guangxi include this two major classes. The method was reliable with good reproducibility. It could be used for the identification and quality control of D.nobile from different regions.
