摘 要:目的探讨定经汤主要药对菟丝子-柴胡调节卵巢早衰的作用机制。方法 SD大鼠随机分为4组:正常大鼠组、模型组、定经汤组和菟丝子-柴胡组;分别检测4组大鼠血清中与卵巢早衰相关的生物分子:β-EP、IL-1、NOS、NO、ER、PR、GnRH、CRH、FSH、LH、E2、P、ACTH和CORT;利用因子分析方法探究菟丝子-柴胡干预下各生物分子之间的相互调控关系。结果菟丝子-柴胡干预状态生殖内分泌网络主调控因子主要包括垂体-卵巢轴4个生物分子、垂体-肾上腺轴靶腺层生物分子CORT以及部分下丘脑生物分子;GnRH是菟丝子-柴胡干预状态主调控因子与其他公共因子共有的生物分子。结论菟丝子-柴胡一方面可以直接调节垂体-肾上腺轴生物分子,另一方面又可以通过下丘脑间接调节垂体-肾上腺轴生物分子;GnRH是各调控因子之间的枢纽生物分子。Objective To explore the biological mechanisms of Dingjing decoction herbs couples of Cuscuta Semen-Bupleuri Radix on the premature ovarian failure( POF). Methods 120 SD rats were randomly divided into four groups including the normal group,the model group,the Dingjing Decoction group and the Cuscuta Semen-Bupleuri Radix group. The biomolecules associated with POF such as β-EP,IL-1,NOS,NO,ER,PR,GnRH,CRH,FSH,LH,E2,P,ACTH and CORT were detected. Regulation relations of biological molecules were explored based on factor analysis.Results The master regulatory factors regulated by Cuscuta Semen-Bupleuri Radix mainly contained the pituitary-ovarian axis four biomolecules,the pituitary-adrenal axis biomolecule CORT and part of hypothalamus biomolecules. GnRH was the common biomolecule in the hypothalamus part of the first and the other common factors regulated by Cuscuta SemenBupleuri Radix. Conclusion Cuscuta Semen-Bupleuri Radix can directly modulate pituitary-adrenal axis molecules,as well as indirectly modulate the pituitary-adrenal axis molecules through hypothalamus. GnRH plays a pivotal role in these factors.
