摘  要:目的探讨水通道蛋白-9(AQP-9)在羊水过少产妇和羊水量正常的孕产妇血清及胎盘组织中的表达差异。方法采用免疫组化方法(SP法)分别检测30例羊水过少和30例羊水量正常的孕产妇胎盘组织中AQP-9的蛋白表达水平,并对其表达强度行半定量分析。同时,采用酶联免疫吸附法(ELISA)测定羊水过少组与对照组外周血AQP-9的浓度水平。结果 AQP-9蛋白在羊水过少组与对照组孕产妇的胎盘中均有阳性表达。羊水过少组胎盘中的AQP-9蛋白表达水平(3.40±1.35)明显比对照组低(5.03±1.74),其表达差异有统计学意义(t=2.32,P<0.05);同时羊水过少组(234.72±67.52)与正常组(243.57±47.58)血清中AQP-9的平均浓度比较差异无统计学意义(t=2.85,P>0.05)。结论羊水过少组孕产妇胎盘中AQP-9蛋白的低表达,提示AQP-9在孕产妇胎盘中的异常表达可能是羊水过少发生的原因之一。Objective To explore the difference of AQP-9 expression in placenta and serum between pregnant women of oligohydramnios and normal pregnant women .Methods Immunohistochemistry was performed to determine the expression of AQP-9 in placenta from 30 cases of oligohydramnios ( oligohydramnios group ) and 30 normal cases ( control gorup ) , and the expression of AQP-9 was analyzed using semi-quantitative method.Meanwhile enzyme linked immunosorbent assay (ELISA) was performed to determine the concentration of AQP-9 in serum of two groups .Results AQP-9 in placenta of two groups was positively expressed .The expression of AQP-9 (3.40 ±1.35) was lower in the oligohydramnios group than in the control group (5.03 ±1.74),and there was significant difference (t=2.32, P〈0.05). Meanwhile there was no significant difference in the expression of AQP-9 in serum between two groups (oligohydramnios group:234.72 ± 67.52, control group:243.57 ±47.58;t=2.85, P〉0.05).Conclusion The low expression of AQP-9 in placenta of pregnant women of oligohydramnios indicates that abnormal expression of AQP-9 might be one of the causes of oligohydramnios .
