内毒素及NO抑制剂对体外培养山羊肝细胞增殖的影响

摘  要:为了研究内毒素(LPS)和NO酶抑制剂(NOSI)[氨基胍(AG)、左旋硝基精氨酸(L-NNA)、NG-硝基-L-精氨酸甲酯(L-NAME)]对肝细胞增殖的影响,试验通过体外培养山羊肝细胞,应用LPS结合NO酶抑制剂对细胞进行处理观察细胞增殖的变化。结果表明:当LPS>1μg/m L时抑制体外肝细胞的增殖,且随着剂量的增加差异显著(P<0.05),说明大剂量的LPS能抑制肝细胞的增殖。应用NOSI(AG,L-NNA,L-NAME)结合1μg/m L LPS处理肝细胞,则发现AG(1 mmol/L)+LPS试验组与LPS对照组肝细胞的增殖差异显著(P<0.05);LPS+L-NAME(1 mmol/L)处理的d组显著高于LPS对照组,差异显著(P<0.05),使吸光度达到最高水平;LPS+L-NNA(1 mmol/L)处理的γ组、δ组与LPS对照组相比差异显著(P<0.05)。说明随着NOSI(AG,L-NNA,L-NAME)浓度的增加,NOSI可以颉颃LPS对肝细胞增殖的抑制,使肝细胞的增殖水平在一定程度上恢复到正常。To study the effects of lipepolysaccharide (LPS) as endotoxin and nitric oxide synthase inhibitors (NOSI) including aminoguanidine (AG) , L - N - nitroarginine ( L - NNA) , NG - nitro - L - arginine methyl ester ( L - NAME) on the proliferation of goat hepatocytes. The goat hepatocytes cultured in vitro, were treated with LPS, or LPS in combination with AG, L - NNA, and L - NAME as NOSI, and then used to observe the changes in the proliferation of goat hepatocytes. The results showed that the proliferation of goat hepatocytcs in vitro was significantly inhibited (P 〈 0.05) by LPS in a dose - dependent manner when the concentration of LPS was higher than 1 μg/mL, indicating that large doses of LPS could inhibit the proliferation of hepatocytes. The goat hepatocytes were treated with AG, L -NNA, and L -NAME in com- bination with LPS of 1 μg/mL, it was found that there was a significant difference ( P 〈0.05 ) on the proliferation of goat hepatocytes between the experimental group with AG of 1 mmol/L in combination with LPS of 1 μg/mL and the control group with LPS ; the d group treated with LPS in combination with L - NAME was significantly higher ( P 〈 0.05 ) that the control group with LPS, which made the absorbance reach the highest level; there were significant differences (P 〈0.05) in the γ and δ groups treated with LPS in combination with L- NAME compared with the control group with LPS. The results indicate that with the increases of the concentrations of NOSI ( AG, L - NNA, and L - NAME) concentrations, NOSI (AG, L- NNA, L -NAME) can antagonize the inhibition of LPS on the proliferation of the hepatocytes, and make the proliferation level of the hepatocytes restore normal to a certain extent.
