摘 要:目的:通过骨灵膏及其拆方制剂对骨关节病(OA)体外培养软骨细胞的作用,探讨其对OA的作用机制。方法:SD大鼠随机分为骨灵膏、骨膏、灵膏、塞来昔布、模型及正常组6组,冷固法建立OA模型,造模成功后,各治疗组给予相应药物灌胃,其余组给予0.9%氯化钠溶液灌胃,给药10周后,用大鼠含药血清对体外传代培养的OA大鼠软骨细胞进行处理;镜下方格法计数细胞密度,免疫荧光检测Ⅱ型胶原的表达,CCK8检测软骨细胞光吸收值,收集各组大鼠脾脏和胸腺计算其指数。结果:4用药组含药血清处理的软骨细胞密度明显高于模型组(P<0.01);骨灵膏与模型组、骨膏组、灵膏组及塞来昔布组比较,软骨细胞的OD显著增加(P<0.01);与正常组比较,模型组大鼠脾脏指数(SI)及胸脾指数(TI)显著降低(P<0.01);骨灵膏组与模型组、骨膏组、灵膏组及塞来昔布组比较大鼠的SI、TI显著增加(P<0.01)。结论:骨灵膏可提升局部软骨细胞增殖活力,阻止OA关节软骨退行性变,其作用明显优于其拆方制剂及塞来昔布。Objective: Study on the effects of Guling Gao (GLG) and its disassembled prescriptions on the cartilage cells in vitro culture of osteoarthritis (OA), to discuss the effect mechanism of OA. Methods: SD rats were randomly divided into normal group (NG) and GLG group, Gu Gao group (GUG), Ling Gao group (LG), celecoxib (CLXB) group, model group (M), the OA model was established by using cold-solid method. After the model was successfully established, the treatment groups were gavaged with the corresponding drugs, the other groups were gavaged with saline. To deal with the OA cartilage cells in vitro subculture by using the drug-containing serum of rats in each group after 10 weeks of administration. The cell density were counted by microscopic squares method, the expression of Ⅱ type collagen were detected by immunofluorescence staining. The light absorption value of cartilage cells were detected by CCK8, the thymus and spleen of the rats in each group were collected to calculate the index. Results: The cartilage cell density was higher than M group significantly after treatment by GLG etc drugcontaining serum (P〈0.01). Compared with M, GUG, LG and CLXB group, the OD of cartilage cells in GLG group was increased (P〈0.01). Compared with NG group, the TI and SI in M group were reduced significantly (P〈0.01). Compared with CLXB and LG, GUG, M group, the TI and SI in GLG group were increased (P〈0.01). Conclusion: GLG could promote local cartilage cell proliferation activity, inhibit OA articular cartilage degeneration, which effect is superior than its disassembled prescriptions and CLXB.
