摘 要:目的:探讨首乌方对美多芭诱发异动症模型大鼠行为学指标及其纹状体细胞外液糖类物质水平的影响。方法:采用6-羟多巴胺(6-OHDA)脑内立体定位注射造成偏侧帕金森病大鼠模型,成模动物分为美多芭组和美多芭+首乌方组,另设假手术组。所有动物连续给药22d,在第1、7、14、21天给药后对大鼠进行异常不自主运动(AIM)评分。第22天采用微透析技术对大鼠纹状体细胞外液进行取样,并采用ISUCSflex分析仪监测样本中葡萄糖、乳酸水平动态变化。结果:与假手术组相比,美多芭组、美多芭+首乌方组AIM评分均显著升高(P<0.05,P<0.01);与美多芭组相比,美多芭+首乌方组AIM评分降低,但差异无统计学意义。第22天,与假手术组相比,美多芭组大鼠纹状体细胞外液葡萄糖、乳酸水平均显著升高(P<0.01),与美多芭组相比,美多芭+首乌方组大鼠纹状体细胞外液葡萄糖、乳酸水平降低(P<0.05)。结论:首乌方配伍美多芭改善异动症模型大鼠行为学症状,其作用机制或与其改善LID大鼠脑内糖类物质代谢障碍有关。Objective: To investigate effects of Shouwu Fang (SWF) on behaviors of madopar-induced dyskinesia model rats, and on striatal extracellular levels of glucose and lactate. Methods: 6-hydroxydopamines were injected to produce Parkinson's disease (PD) rat model. Model rats were daily given Madopar, or Madopar plus SWF. The sham-operated rats were given water. All rats were treated for 22 days continuously. On 1st, 7th, 14th, and 21st day of the treatment, abnormal involuntary movements (AIMs) were measured. On 22nd day, in vivo microdialysis was used to collect striatal extracellular fluid. All samples were analyzed by ISUCS^ftex. Results: AIM scores of Madopar- and Madopar plus SWF-treated rats were higher than the sham-operated rats (P〈0.05, P〈0.01). Madopar plus SWF-treated rats had lower AIM scores than Madopar-treated rats, there was no significant difference. On 22nd day, in 6-OHDA-lesioned rats, striatal levels of glucose and lactate increased (P〈0.01) than sham group. Levels of glucose and lactate in Madopar plus SWF-treated rats were lower than Madopar-treated rats (P〈0.05). Conclusion: In 22-day treatment, as SWF was combined, behavioral symptoms were improved in dyskinesia model rats. Effects of SWF might be due to its improvement on cerebral carbohydrate metabolism.
