循经取穴针刺对心肌缺血大鼠模型心肌细胞瞬时外向钾通道基因表达的影响

摘　　要：目的:探讨针刺不同经脉腧穴对心肌细胞瞬时外向钾通道(Kv1.4、Kv4.2、Kv4.3)及其辅助亚基(KCh IP2)表达水平变化的影响。方法:通过大剂量注射盐酸异丙肾上腺素(ISO)建立心肌缺血大鼠模型,随机分为5组:对照组、模型组、内关组、列缺组和非经非穴组。针刺干预7d后,采用实时荧光定量聚合酶链式反应(Real-Time PCR)技术,检测大鼠左心室肌上述基因的表达情况。结果:与模型组比较,内关组、列缺组待测基因表达显著升高,差异有统计学意义(P<0.05);与内关组比较,列缺组、非经非穴组待测基因表达显著降低,差异有统计学意义(P<0.05);结论:心肌细胞瞬时外向钾通道(Kv1.4、Kv4.2、Kv4.3)及其辅助亚基(KCh IP2)的异常表达是心肌缺血的发病机制之一,针刺手厥阴心包经内关穴抗心肌缺血具有循经特异性。Objective： To observe the effect of needling different acupoints on the expression of myocardial transient outward potassium channel （Kvl.4, Kv4.2, Kv4.3） and auxiliary subunit （KChlP2）. Methods： The model of myocardial ischemia was established by injecting large doses of Isoprenaline Hydrochloride （ISO）. And then the rats were randomly divided into five groups： control group, model group, Neiguan （PC6） group, Lieque （LU7） group and non-acupoint group. Electro-acupuncture stimulation was given to the corresponding acupoints for 7 days, then the expression level of these genes on the left ventricular was detected by real-time fluorescence quantitative polymerase chain reaction （Real-time PCR）. Results： Compared with the model group, the genes expression in PC6 and LU7 group increased significantly, and the difference was statistically significant （P〈0.05）. Compared with PC6 group, the expression level of these genes in LU7 group and non-acupoint group decreased significantly, and the difference was statistically significant （P〈0.05）. Conclusion： The changes of the expression of myocardial transient outward potassium channel （Kvl.4, Kv4.2, Kv4.3） and auxiliary subunit （KChlP2） might be one of the pathogenesis of myocardial ischemia, and acupuncture PC6 of hand-Jueyin pericardium meridian had its own unique characteristic in treating diseases.
