摘 要:目的:观察电针四关穴对抑郁大鼠海马星形胶质细胞形态的影响;方法:成年雌性SD大鼠24只,随机分为空白组、模型组、电针组,每组8只,空白组不予任何处理,模型组采用慢性不可预见性温和刺激联合孤养造模法建立抑郁大鼠模型8周,电针组于造模第5周后行电针刺四关穴治疗。8周后3组大鼠分别行心脏灌注,取海马组织于光镜和电镜下进行观察。结果:与空白组比较,模型组大鼠海马星形胶质细胞形态和超微结构受损,光镜下观察主要表现为细胞核的固缩,电镜下观察主要表现为胞质中粗面内质网明显扩张和线粒体的嵴疏松变;与模型组大鼠比较,电针组大鼠海马星形胶质细胞上述表现明显减轻。结论:电针四关穴可以有效修复慢性应激致抑郁大鼠海马星形胶质细胞形态和超微结构损伤,这可能是电针抗抑郁的重要机制之一。Objective: To observe the influence of electro-acupuncture at si-guan [Hegu (LI4) and Taichong (LR3)] on the morphology of hippocampal astrocytes in depressive rats. Methods: Twenty-four at adult female SD rats (220-250g, 7-8 weeks) were randomly divided into normal control group (NC, n=8), depression model group (DM, n=8) and electro-acupuncture group (EA, n=8). The rats were not given any processing in NC group. The rats were produced by chronic unpredictable mild stress and separation for 8 weeks to build depression model rat in DM group. The rats were given electro-acupuncture at si-guan treatment in the EA group on 5th week after model made. After 8 weeks, heart perfusion was operated to obtain specimens, the hippocampal tissues were got and observed under light microscope and electron microscope. Results: Compared with NC group, rats in DM group suffered morphological changes (mainly for the nucleus pycnosis) and ultrastructure damage (mainly for the cytoplasm rough endoplasmic reticulum expansion and mitochondrial cristae loose change) in hippocampal astrocytes. Compared with DM group, the rats in EA group had significant relieved morphology of hippocampal astrocytes. Conclusion: Electro-acupuncture at si- guan could effectively relieve the morphological damage and ultrastructure damage of chronic stress in hippocampal astrocytes of rats, and it might be one of the most important mechanisms of antidepressant.
