摘 要:目的:对附子在急性心力衰竭模型下的入血成分进行分析,以期阐明附子治疗急性心力衰竭的有效成分及作用机制。方法:文章用戊巴比妥钠建立大鼠急性心力衰竭模型,UPLC/MS技术比较分析空白血清、给药血清及附子提取液的差异,根据保留时间和分子量,确认附子在急性心力衰竭模型下的原型入血成分,并进行体内动态变化的初步研究。结果:其中12个以原型入血。尼奥灵、异翠雀碱、次乌头碱在5-30min的时间内始终存在于大鼠血清中,且峰面积在10min出现首次达峰,此后亦有明显上升趋势。准格尔乌头碱、新乌头碱也始终存在于大鼠血清中,但其峰面积变化不显著。其他成分峰面积相对较小且变化不明显。结论:这些血中移行成分可能是附子在体内的直接作用物质,对其进行深入的研究将有助于阐明附子治疗急性心力衰竭的有效成分及作用机制。Objective: To analyze the effective components in plasma and dynamic changes of prepared common monkshood branched root in the rat model of acute heart failure, in order to explain the effective components and action mechanism of prepared common monkshood branched root in treating with acute heart failure. Methods: Rat models of acute heart failure were established by using mebumalnatrium. UPLCfMS method was used to compare and analysis the differences among serum of rats in control group, serum of rats in treatment group and extract of prepared common monkshood branched root, in order to verify the components in plasma of prepared common monkshood branched root in the rat model of acute heart failure according to the retention time and molecular weight, and preliminary study about the dynamic changes of effective components in vivo. Results: 20 alkaloids components were detected in prepared common monkshood branched root, and 12 of them entered into blood with unchanged metabolite. Neoline, isodelphinine, hypaconitine always existed in serum of rats in the 5-30 min, and peak area for the first time in the peak at the first 10 minites, since then also had obvious rising trend. Zhungeer aconitine and new aconitine also always existed in serum of rats, but the peak area change was not significant. The peak area of other components was relatively small and the change was not obvious. Conclusion: These transitional components in blood might be the material of direct effect of prepared common monkshood branched root in vivo, and made a deep research on it would contribute to explain the active components and action mechanism of prepared common monkshood branched root in treating with acute heart failure.
