摘  要:探讨当归多糖对麻黄素小鼠肝组织抗氧化酶活性和核转录因子-κB(NF-κB)、肿瘤坏死因子(TNF-α)表达的影响及其可能作用机制.小鼠连续腹腔注射4.0g·L-1麻黄素溶液,同时用当归多糖溶液(12.5,25.0g·L-1)进行灌胃,分别在5,10,15d时用分光光度法检测各组小鼠肝组织T-AOC、GSH-PX活性的变化,免疫组化法检测小鼠肝组织中NF-κB和TNF-α表达的变化.结果显示实验对照组小鼠肝组织T-AOC、GSH-PX活性明显降低(P<0.01),肝组织NF-κB和TNF-α表达量显著增强(P<0.05或P<0.01),而当归多糖组T-AOC、GSH-PX活性与实验对照组相比显著升高,NF-κB和TNF-α表达量明显降低(P<0.05或P<0.01).结果表明,麻黄素影响小鼠肝组织抗氧化酶活性和NF-κB、TNF-α的表达,当归多糖能够提高小鼠肝组织抗氧化酶活性,抑制NF-κB和TNF-α的表达,对麻黄素致小鼠肝损伤有一定的保护作用.The effect of Angelica sinensis polysaccharide on activities of antioxidant enzyme and expressions of NF‐κB、 TNF‐αin liver tissue of the mice injured by ephedrine are investigated , and its possible mechanism is also explored . The mice are intraperitoneal injected 4.0 g·L^ -1 ephedrine , and irrigated to stomach with A ngelica sinensis polysaccharide solution (12.5 ,25.0 g·L^ -1 ) after injected ephedrine 1 h later . The samples at 5 ,10 and 15 d respectively ,activities of hepatic histological T‐AOC、GSH‐PX are detected by spectrophotometer . The expressions of NF‐κB and TNF‐αare measured by immunohistochemistry in liver of mice at different developmental stages . Results show the activities of hepatic histological T‐AOC、GSH‐PX of experiment control group significantly reduce (P〈 0.01) , the positive expressions of NF‐κB and TNF‐αof live obviously increase(P〈0.05 or P〈0.01);Compared with experiment control group , the activities of T‐AOC、 GSH‐PX increase significantly and the positive expressions of NF‐κB and TNF‐αof liver significantly reduce of Angelica sinensis polysaccharide group(P 〈0.05 or P〈 0.01 ) . Indicate A ngelica sinensis polysaccharide can improve liver tissue antioxidant capacity and inhibiting the expressions of NF‐κB and TNF‐α, which may be one of the important mechanisms for the treatment of liver injury by ephedrine .
