白三烯B4、D4、基质金属蛋白酶2在动脉粥样硬化大鼠中的表达

摘  要:目的观察白三烯B4(LTB4)、白三烯D4(LTD4)、基质金属蛋白酶2(MMP2)在动脉粥样硬化大鼠中的表达,探讨其致动脉粥样硬化的机制。方法选取雄性SD大鼠24只,随机分为正常组和动脉硬化组。正常组给予普通饲料喂养,动脉硬化组给予高脂饲料喂养,14周后建立动脉粥样硬化大鼠模型,处死大鼠,采血,检测血脂,ELISA法检测血清和主动脉中白LTB4、D4、MMP2的含量。结果与正常组比较,动脉硬化组甘油三酯、胆固醇、低密度脂蛋白水平明显升高(P<0.05),高密度脂蛋白水平减低(P<0.05)。动脉硬化组主动脉以及血液中LTB4、LTD4、MMP2的含量与正常组比较明显升高(P<0.05)。结论 LTB4、D4、MMP2在动脉硬化大鼠血液及主动脉中均异常表达,是致动脉粥样硬化重要的炎症因子。Objective To observe the expression of leukotrieneB4(LTB4), leukotriene D4(LTD4)and Matrix metalloproteinas?es2(MMP2)in atherosclerotic rats and explore the mechanism of atherosclerosis. Methods Twenty-four male SD rats were divided into normal group and atherosclerotic group. Normal group ratswere fed with normal diet,Atherosclerotic group rats were fed withhigh-fat di?et. After 14 weeks, atherosclerosis models were found, thenthe rats were killed and the lipids,LTB4,LTD4 and MMP2 in the blood and aorta were measure by Enzyme-Linked Immunosorbnent Assay. Results Compared with normal group,there was significantly increased in the level of Triglyceride (TG), cholesterol (CHOL), low density lipoprotein (LDL) in atherosclerotic group(P〈0. 05)and the level of high-density lipoprotein (HDL) was lesser(P〈0. 05). Meanwhile,the expression of LTB4、LTD4、MMP2 in the blood and artor of atherosclerotic rats were significantly higher than those in normal group(P〈0. 05). Conclusion The expression of LTB4,LTD4,MMP2 are abnormal in the blood and artor of atherosclerotic rats,which are important inflammatory factors that bring aboutatherosclerosis.
