摘  要:目的研究黄芪、水蛭及其配方含药血清干预系膜增生性肾小球肾炎的可能作用机制。方法 20只SD大鼠随机分为黄芪高血清组、水蛭高血清组、黄芪高水蛭高血清组、黄芪低水蛭高血清组,每组5只。大鼠给药量5 g/(kg·d)为低浓度,2倍剂量为高浓度,各组均配成0.5 g/ml混悬液,每天早晚各灌胃1次,连续灌胃5天,末次灌胃1.5 h后制备含药血清。检测各组不同浓度(10%、20%、40%、80%)的含药血清对大鼠系膜细胞增殖的影响,计算细胞存活率,筛选出各组最佳浓度。系膜细胞以1×105个/孔接种于6孔板中,每孔2 ml,分为正常组、模型组、黄芪高血清组、水蛭高血清组、黄芪高水蛭高血清组、黄芪低水蛭高血清组,除正常组外其余各组加入脂多糖诱导大鼠肾小球系膜细胞增殖,再分别加入筛选出的不同浓度的含药血清进行干预,检测各组系膜细胞中核转录因子(NF-кB)、基质金属蛋白酶2(MMP-2)、Fas、Fas L mRNA转录水平。结果黄芪高血清组、水蛭高血清组、黄芪高水蛭高血清组、黄芪低水蛭高血清组含药血清最佳浓度分别为10%、20%、40%、20%。与正常组比较,模型组系膜细胞中NF-кB、MMP-2、Fas、Fas L mRNA表达明显升高(P<0.05),而各含药血清组NF-кB、MMP-2、Fas、Fas L mRNA表达较模型组显著降低(P<0.01)。结论黄芪、水蛭及其配方含药血清可抑制大鼠系膜细胞增殖,其作用机制可能与下调NF-кB、MMP-2、Fas、Fas L mRNA表达水平有关。Objective To study the possible mechanism of containing serum of Huangqi [Astragalus membranaceus( Fisch.) Bunge],) and their formulation on mesangial proliferative glomerulonephritis( Ms PGN). Methods Twenty SD rats were randomly assigned to high-dose Huangqi serum group,high-dose Shuizhi serum group,high-dose Huangqi and Shuizhi serum group,low-dose Huangqi and high-dose Shuizhi serum group with 5 rats in each group.The concentration of 5 g /( kg·d) was defined as low-dose,while 10 g /( kg·d) as high-dose. Suspension of 0. 5g / ml was prepared to apply on the mornings and evenings by gavage everyday for 5 days. Drug-containing serum was prepared 1. 5 h after the last gavage. Influence on cell proliferation was investigated in different concentration( 10%,20%,40%,80%) of drug-containing serums and cell survival rate was calculated to screen the concentration with best result in each group. Mesangial cells were planted in 6-well plates with 1 × 105cells( 2 ml) per well.They were divided into normal group,model group,high-dose Huangqi serum group,high-dose Shuizhi serum group,high-dose Huangqi and Shuizhi serum group,low-dose Huangqi and high-dose Shuizhi serum group. Except for the normal group,all other groups were stimulated with lipopolysaccharide( LPS) to induce mesangial cells into proliferation. Then the screened drug-containing serum with different concentration was added and the transcription levels of nuclear factor( NF-кB),matrix metalloproteinase-2( MMP-2),Fas,Fas L mRNA were detected in each group.Results Best results achieved in high-dose Huangqi serum group,high-dose Shuizhi serum group,high-dose Huangqi and Shuizhi serum group,low-dose Huangqi and high-dose Shuizhi serum group were with the concentration of 10%,20%,40%,20%,respectively. Compared with the normal group,the expression of NF-кB,MMP-2,Fas,Fas L mRNA in the model group was significantly increased( P〈0. 05).
