胃炎饮对胃溃疡模型大鼠溃疡组织生长因子表达的影响

摘　　要：目的探讨胃炎饮促进胃溃疡愈合的可能作用机制。方法 40只Wistar大鼠随机分为假手术组、模型组、胃炎饮高剂量组、胃炎饮低剂量组、奥美拉唑组,每组8只。除假手术组外,其余各组大鼠采用冰醋酸腐蚀法建立胃溃疡大鼠模型,造模3天后,胃炎饮高、低剂量组大鼠分别灌服浓度为2.14、1.07 g/ml的胃炎饮方药液10 ml/kg,奥美拉唑组灌服0.368 mg/ml的奥美拉唑药液10 ml/kg,假手术组、模型组灌服等量生理盐水。治疗14天后,肉眼观察胃壁冰醋酸腐蚀处大体形态,测量、计算胃黏膜溃疡面积及溃疡抑制率,检测胃黏膜表皮生长因子(EGF)、血管内皮生长因子(VEGF)及其mRNA表达。结果模型组、胃炎饮高、低剂量组、奥美拉唑组大鼠胃溃疡面积分别为(26.8±9.99)mm2、(10.0±4.62)mm2(19.5±6.02)mm2、(17.5±6.18)mm2,各用药组溃疡面积均较模型组显著缩小(P<0.05或P<0.01),且胃炎饮高剂量组溃疡面积明显小于胃炎饮低剂量组与奥美拉唑组(P<0.05)。与假手术组比较,模型组大鼠胃黏膜EGF、VEGF表达的平均光度值显著升高,平均灰度值显著下降(P<0.05或P<0.01);各治疗组与模型组比较,其平均光度值显著升高,平均灰度值显著下降(P<0.05或P<0.01),且胃炎饮高剂量组改善程度优于胃炎饮低剂量组与奥美拉唑组(P<0.05或P<0.01)。与假手术组比较,模型组大鼠胃黏膜EGF mRNA、VEGF mRNA表达显著升高(P<0.05或P<0.01);各治疗组与模型组比较,EGF mRNA、VEGF mRNA表达显著升高,且胃炎饮高剂量组显著高于奥美拉唑组与胃炎饮低剂量组(P<0.05或P<0.01)。结论胃炎饮可能通过提高胃溃疡模型大鼠胃黏膜生长因子的表达,从而促进胃黏膜修复及溃疡愈合。Objective To discuss the possible mechanisms of ulcer healing process with Weiyan Yin（ decoction for gastritis）. Methods Totally 40 Wistar rats were randomly assigned to sham-operation group,model group,Weiyan Yin high-dose group,Weiyan Yin low-dose group and omeprazole group with 8 in each group. Except for sham-operation group,acetic acid corrosion method was adopted to make gastric ulcer rat models. After 3 days,Weiyan Yin high-dose group and Weiyan Yin low-dose group were given Weiyan Yin 10 ml / kg by the concentration of 2. 14,1. 07 g / ml,while omeprazole group was given omeprazole liquid 10 ml / kg by the concentration of 0. 368 mg / ml.Sham-operation group and model group were given the same amount of normal saline. After the 14-day treatment,the ulcer condition was observed by naked eyes. The area of the gastric ulcer was measured and the inhibiting rate of ulcer was calculated. The expression of epidermal growth factor（ EGF）,vascular endothelial growth factor（ VEGF） and VEGF mRNA was detected. Results The ulcer area for the model group,Weiyan Yin high-dose group,Weiyan Yin low-dose group,omeprazole group was（ 26. 8 ± 9. 99） mm2,（ 10. 0 ± 4. 62） mm2,（ 19. 5 ± 6. 02） mm2,（ 17. 5 ±6. 18） mm2,respectively. Ulcer area in all treatment groups was significantly reduced compared with the model group（ P〈0. 05 or P〈0. 01） and ulcer area of Weiyan Yin high-dose group was smaller than that of Weiyan Yin low-dose group and omeprazole group（ P〈0. 05）. Compared to the sham-operation group,the average luminasity value of EGF and VEGF expression in the model group was significantly higher,while the average gray value was lower（ P〈0. 05 or P〈0. 01）. The average luminasity value was significantly higher in all treatment groups than in the model group,while the average gray value was lower（ P〈0. 05 or P〈0. 01）.
