摘 要:传统的湖泊沉积物硅藻提纯方法存在周期较长、过筛会损失某些特定硅藻种类等问题,从而造成硅藻群落分析出现偏差。基于硅藻提纯的实验原理和传统沉积物硅藻提纯技术,选取广东三个典型水库的沉积物的五个不同样品,设置了三组优化实验,建立了一套适合水库沉积物硅藻的提纯方法。实验表明,该方法能有效地提纯和富集水库沉积物中的硅藻,在简化实验操作的同时,明显缩短了提纯时间,并且更有利于其后硅藻壳体种属的鉴定,在水库沉积物硅藻群落分析具有较好的应用价值。Traditional methods for extraction and concentration of diatoms from lake sediments have been more and more criticized for being time consuming and losing particular species during sieving, and therefore resulting in biased diatom assemblages. Based on the principles and procedures of traditional concentration methods, an improved method was developed through three sets of optimization test on 5 samples from three different reservoirs of Guangdong Province. The experiment results showed that this improved method can efficiently extract and concentrate diatoms from sediments, and meanwhile has the advantages such as simplifying experiment procedure, shortening experiment time, and facilitating diatom identification. Thus, this method has great potential in studying diatom paleoecology with reservoir sediments.
