调更汤对围绝经期雌性大鼠生殖内分泌影响的实验研究

摘　　要：目的观察调更汤对围绝经期雌性大鼠生殖内分泌的影响,探讨其对围绝经期综合征症状改善的作用机制。方法 40只4～5月龄雌性SD大鼠,随机选择10只为青年组,其余30只均摘除双侧卵巢去势。随机分为3组,分别为模型组、中药组、西药组。围绝经期模型组与青年组予0.9%Na Cl溶液灌胃,中药组予调更汤,西药组予倍美力。灌药8周后,检测下丘脑及血清促卵泡刺激素（FSH）、促黄体生成素（LH）、雌二醇（E2）、睾酮（T）、瘦素（Leptin）水平,观察各组大鼠生殖内分泌情况。结果 ①造模成功后血清及下丘脑性激素的变化：与青年组比较,模型组血清E2、T水平显著降低（P〈0.01）,FSH、LH、Leptin水平显著升高（P〈0.05,P〈0.01）。②中药组给予调更汤后血清性激素的变化：与模型组比较,中药组血清E2水平显著提高（P〈0.05）,FSH、Leptin水平显著降低（P〈0.05,P〈0.01）,但对LH、T影响不明显（P〉0.05）。与西药组比较,中药组血清E2、T含量降低,差异有统计学意义（P〈0.05,P〈0.01）。③中药组给予调更汤后下丘脑性激素的变化：与模型组比较,中药组下丘脑E2、FSH、LH、T变化不明显（P〉0.05）,但可明显降低下丘脑Leptin含量（P〈0.05）。与西药组比较,中药组下丘脑E2、T含量降低,差异有统计学意义（P〈0.01）。结论调更汤可通过提高血清E2水平,降低FSH与Leptin水平,进而减缓卵巢退行性变,延缓卵巢衰退,提高卵巢功能,从而起到治疗围绝经期综合征的作用。Objective To discuss the clinical efficacy of＂Tiaogeng Decoction＂on reproductive endocrine of climacteric rats. Methods Forty female SD rats aged 4 ～5 months were randomly divided into four groups. Ten for youth group and the other 30 were removed bilateral ovaries castration and then were randomly divided into model group,Chinese medicine group and the western medicine group. The model group and the youth group were given gavages with 0. 9% Na Cl solution; Chinese medicine group were given ＂Tiaogeng Decoction ＂; western medicine group were given Premarin. After 8 weeks,hypothalamus of rats,separating serum were collected,and the levels of LH,FSH,E2,T and Leptin were examined,and the reproductive endocrine in each group were observed. Results ①After molding,the change of serum sex hormone in hypothalamus： the content of serum E2、T in the model group significantly decreased compared with the youth group（ P〈0. 01）,while the levels of FSH,LH and Leptin significantly increased（ P〈0. 05,P〈0. 01）. ② After given ＂Tiaogeng Decoction＂,the change of serum sex hormone in the Chinese medicine group： the level of serum E2 in the Chinese medicine group significantly increased compared with the model group（ P〈0. 05）,while the content of FSH and Leptin significantly decreased（ P〈0. 05,P〈0. 01）,but the levels of LH and T did not change greatly（ P〉0. 05）. ③ After given ＂Tiaogeng Decoction＂,the change of hypothalamic sex hormone in the Chinese medicine group： levels of hypothalamic E2,FSH,LH and T did not change greatly（ P〈0. 05）compared with the model group,but the content of hypothalamic Leptin significantly decreased（ P〈0. 05）. Compared with the western medicine group. the levels of hypothalamic E2 and T decreased significantly in the Chinese medicine group（ P〈0. 05）. Conclusion＂Tiaogeng Decoction＂can raise the serum E2 level in climacteric SD rats effectively,decrease the levels of FSH and Leptin,slowi down ovary retrogression and ovary rece
